摘要:为了探讨炙黄芪与炙红芪干预脾气虚证的药效差异,将110只SD雄性大鼠随机分为10只空白组与100只造模组,造模成功后将造模组分为模型组,补中益气丸组,炙红芪高、中、低剂量组以及炙黄芪高、中、低剂量组,每组10只。空白组正常饲养,模型组持续施加造模条件,补中益气丸组每天灌胃给予18.9g·kg^(-1)补中益气丸水溶液,炙黄芪与炙红芪高、中、低剂量组每天分别灌胃给予18.9、12.6、6.3g·kg^(-1)对应剂量炙黄芪与炙红芪水煎液。给药组每天早晨以10m L·kg^(-1)给予相应药物,空白组与模型组给予同等剂量蒸馏水,每天1次,连续给药15d。药物干预完成后,计算各组大鼠脾脏指数和胸腺指数,对脾脏组织切片,HE染色,并观察病变情况;检测各组大鼠血常规、D-木糖(D-xylose)、胃动素(MTL)、白细胞介素-2(IL-2)、干扰物-γ(IFN-γ)、免疫球蛋白A(IgA)及胃黏膜胃蛋白酶(pepsin)等指标水平。结果显示,与空白组相比,模型组大鼠血常规各项指标、脾脏指数、胸腺指数、D-xylose、IFN-γ、IgA、MTL及pepsin水平均明显下降(P<0.01),IL-2水平明显上升(P<0.01),脾脏病理切片结果显示脾脏病变严重。与模型组相比,各给药组的IL-2水平均明显下降(P<0.05或P<0.01);除炙红芪低剂量组对脾脏指数、胸腺指数、IgA水平差异无统计学意义,炙黄芪中剂量组对脾脏指数差异无统计学意义,以及炙黄芪低剂量组对脾脏指数、胸腺指数、IgA、MTL水平的影响在统计学上没有差异外,其余各组血常规各项指标、脾脏指数、胸腺指数、D-xylose、IFN-γ、IgA、MTL及Pepsin水平均明显上升,具有统计学意义(P<0.05或P<0.01),各给药组脾脏病变不同程度的恢复。炙红芪各剂量组与炙黄芪各剂量组相比,炙红芪高、中剂量组对血常规白细胞计数(WBC)水平影响均强于炙黄芪对应剂量组(P<0.05),炙红芪高剂量组对IL-2水平影响及炙红芪高、中剂量组对IFN-γ水平影响均高于炙黄芪相应剂量组(P<0.05或P<0.01),炙红芪各对应剂量组对D-xylose及MTL水平影响均高于炙黄芪相应剂量组(P<0.05或P<0.01),其余指标两药之间无差异;同时,通过比较发现,炙红芪与炙黄芪低剂量组(6.3g·kg^(-1))对IL-2及IFN-γ水平恢复并无差异,而炙红芪高剂量组(18.9g·kg^(-1))明显优于炙黄芪高剂量组(IL-2,P<0.01;IFN-γ,P<0.05)。综上,炙黄芪与炙红芪对脾气虚大鼠均有一定治疗作用。免疫调节方面,当给药剂量为6.3g·kg^(-1)以内时,两药免疫调节能力相当;当给药量超过18.9g·kg^(-1)时,炙红芪免疫调节能力明显优于炙黄芪;消化吸收方面,炙红芪消化吸收作用优于炙黄芪。炙黄芪与炙红芪两药在免疫调节与消化吸收作用的差异,有可能是临床用药有别的原因。
