摘要:目的观察当归养颜祛斑面膜的皮肤毒性,评价其安全性。方法通过皮肤急性毒性试验、长期毒性试验、刺激性试验、变态反应试验,观察当归养颜祛斑面膜对豚鼠或新西兰大耳白兔皮肤的急性毒性反应、长期毒性反应、刺激性及变态反应性。结果皮肤急性毒性试验结果显示,各组豚鼠进食量未见明显减少,呼吸、体态、眼、精神状态、四肢活动、粪便性状均无明显改变,未出现死亡;与空白组比较,各给药组不同时间点豚鼠体质量无明显变化,差异均无统计学意义(P>0.05)。皮肤长期毒性试验结果显示,与空白组比较,中药面膜组豚鼠血液生化学指标[白细胞(WBC)、红细胞(RBC)、血红蛋白(Hb)、血小板(PLT)、谷草转氨酶(AST)、谷丙转氨酶(ALT)、总胆红素(TBIL)、尿素氮(BUN)、肌酐(Cr)]水平无明显变化,差异均无统计学意义(P>0.05);各给药组豚鼠胰、肺、胃、脑皮层、海马CA1区未发生明显病理改变。皮肤刺激性试验结果显示,当归养颜祛斑面膜对新西兰大耳白兔完整及破损皮肤均无皮肤反应及刺激性。皮肤变态反应试验结果显示,与空白组比较,中药面膜组各个时间点变态反应发生率及致敏强度无明显变化,差异均无统计学意义(P>0.05)。结论当归养颜祛斑中药面膜对实验动物无皮肤毒性、刺激性,无变态反应,安全性高,具有良好的应用前景。
Objective To observe the skin toxicity of Angelica sinensis skin-nourishing and freckle-removing mask and evaluate its safety.Methods Acute toxicity test,long-term toxicity test,irritation test and allergy test were conducted to observe the acute toxicity reaction,long-term toxicity reaction,irritation reaction and allergy reaction of the skin of guinea pig or New Zealand white rabbit.Results The results of skin acute toxicity test showed that there was no significant decrease in food intake,no significant change in respiration,physical state,eyes,mental state,limb activity,fecal characteristics,and no death.Compared with the blank group,there were no significant changes in body mass of guinea pig at different time points in each treatment group(P>0.05).Long-term skin to-xicity test results showed that compared with the blank group,there were no significant changes in blood biochemical indexes of guinea pig[white blood cells(WBC),red blood cells(RBC),hemoglobin(Hb),platelets(PLT),aspartate aminotransferase(AST),alanine aminotransferase(ALT),total bilirubin(TBIL),urea nitrogen(BUN),creatinine(Cr)]in the treatment group.There were no significant differences(P>0.05).And there were no significant changes in pancreas,lung,stomach,cerebral cortex and hippocampus CA1 area of guinea pig in each treatment group.The results of skin irritation test showed that Angelica sinensis skin-nourishing and freckle-removing mask had no skin reaction and irritation on both intact and damaged skin of New Zealand white rabbit.The results of skin allergy test showed that compared with the blank group,the incidence of allergy and sensitization intensity of treatment group had no significant changes at each time point,and the differences were not statistically significant(P>0.05).Conclusion Angelica sinensis skin-nourishing and freckle-removing mask has high safety without skin toxicity,irritation,allergy on experimental animals,and it has a good application prospect.
