摘要:目的建立铁棒锤不定根的诱导及增殖培养体系。方法通过组织培养的方法,考察不同诱导方法对铁棒锤不定根诱导及增殖情况的影响。结果 MS+吲哚-3-丁酸(IBA)2.5 mg·L-1+6-苄基腺嘌呤(6-BA)0.5 mg·L-1+聚乙烯吡咯烷酮(PVP)1.0 g·L-1为最适愈伤组织诱导培养基,MS+IBA 0.5 mg·L-1+PVP 1.0 g·L-1为最适不定根诱导培养基,MS+IBA0.5 mg·L-1+PVP 1.0 g·L-1为最适不定根增殖培养基。结论基本建立了铁棒锤不定根的诱导及增殖培养体系,为铁棒锤深层次的资源开发及利用提供了一种新途径。
OBJECTIVE To establish the induction and culture system of adventitious roots of Aconitum pendulum Busch. METHODS By tissue culture, the influences of different explants and plant growth substances on the induction and proliferation of adventitious roots of Aconitum pendulum Busch were investigated. RESULTS Mixture of MS + 2.5 mg·L^-1 indole-3- butyric acid(IBA) + 0.5 mg·L^-1 6-benzylaminopuriue(6-BA) + 1.0 g·L^-1 polyvinyl pyrrolidone(PVP) was the best callus induction medium. Mixture of MS +0.5 mg·L^-1 IBA + 1.0 g·L^-1 PVP was the optimum adventitious root induction medium. Mixture of MS + 0.5 mg·L^-1 IBA + 1.0 g·L^-1 PVP was the most suitable adventitious root proliferation medium. CONCLUSION A culture system is preliminarily established for the induction and proliferation of adventitious roots of Aconitum pendulum, which provides a new way for the resource exploitation and utilization of Aconitum pendulum Busch.
