摘要:目的制备红芪多糖HG-4,测定其理化性质,比较红芪多糖HG-4与红芪总多糖、红芪多糖HG-2、HG-3的微观结构及抗氧化活性。方法采用葡聚糖凝胶(Sephadex) G-75制备红芪多糖HG-4,分别采用苯酚-硫酸法、聚丙烯酰胺凝胶电泳(PAGE)、凝胶渗透色谱(GPC)法测定其含量、纯度、分子量;采用薄层色谱(TLC)法、气相色谱-质谱联用(GC-MS)、红外光谱(IR)、扫描电镜(SEM)测定其单糖组成及结构;采用1,1-二苯基-2-苦基苯肼(DPPH)自由基体外抗氧化试验测定其抗氧化能力。结果测得红芪多糖HG-4含量为96.87%,重均分子量(Mw)为32 000 Da;组成分析结果显示,红芪多糖HG-4由葡萄糖、阿拉伯糖、半乳糖3种单糖组成,摩尔比为4.8∶0.4∶0.5;IR结果显示,HG-4为β-吡喃型多糖;SEM结果显示,红芪多糖HG-2、HG-3、HG-4形成稳定的紧密结构;体外抗氧化试验结果显示,红芪多糖HG-2、HG-3、HG-4均具有抗氧化活性,相较于红芪总多糖HG-2、HG-3、HG-4在低浓度时即可对DPPH自由基发挥作用。结论经Sephadex G-75分离制备的红芪多糖HG-4是一种分子量为32 000 Da,由3种单糖组成,具有抗氧化活性的高纯度多糖,可为后期不同分子量红芪多糖的活性研究提供参考。
Objective To prepare Hedysari radix polysaccharides HG-4,determine its physicochemical property,and make comparisons in microstructure and antioxidant activity among Hedysari radix polysaccharides HG-4,total Hedysari radix polysaccharides,Hedysari radix polysaccharides HG-2 and Hedysari radix polysaccharides HG-3. Methods Hedysari radix polysaccharides HG-4 was prepared by Sephadex G-75. The content,purity and molecular weight thereof were determined by phenol-sulfuric acid,polyacrylamide gel electrophoresis( PAGE) and gel permeation chromatography( GPC),respectively;The composition and structure of monosaccharides were determined by thin-layer chromatography( TLC),gas chromatography-mass spectrometer( GC-MS),infrared spectrum( IRS) and scanning electron microscope( SEM). The antioxidant capacity was determined by 1,1-diphenyl-2-picrylhydrazyl( DPPH) free radical in vitro antioxidant test. Results The content of Hedysari radix polysaccharides HG-4 was96.87%,and the weight average molecular weight( Mw) was 32000 Da. The results of composition analysis showed that Hedysari radix polysaccharides HG-4 consisted of 3 monosaccharides,namely glucose( Glc),arabinose( Ara)and galactose( Gal),molar ratio was 4.8 ∶ 0.4 ∶ 0.5;IR showed that HG-4 was β-pyran type polysaccharides;SEM showed that Hedysari radix polysaccharides HG-2,HG-3 and HG-4 all formed compact and stable structures;in vitro antioxidant tests showed that Hedysari radix polysaccharides HG-2,HG-3 and HG-4 had antioxidant activities and they could exert effects on DPPH free radicals at low concentrations compared with total Hedysari radix polysaccharides. Conclusion Hedysari radix polysaccharides HG-4 isolated by Sephadex G-75 is a high purity polysaccharide with molecular weight of 32 000 Da,which consists of three monosaccharides and has antioxidant activity. It can provide references for the later study on the activity of Hedysari radix polysaccharides with different molecular weight.
