摘要:目的:建立高效液相色谱(HPLC)法同时检测黄花菜中槲皮素、山奈酚、异鼠李素、β-谷甾醇4种成分,探索合理开发利用黄花菜的科学依据。方法:采用安捷伦C18色谱柱(250mm×4.6mm,5μm),乙腈-0.1%磷酸水作为流动相,梯度洗脱,流速:1m L/min,柱温:30℃,检测波长:210nm。运用SPSS软件对结果进行分析。结果:槲皮素、山奈酚、异鼠李素、β-谷甾醇分别在27.96-295.18、27.78-304.64、19.09-181.10、3.86-55.43μg与峰面积良好的线性关系(r>0.9992);4种成分的平均回收率为99.25%、99.06%、98.80%、96.91%,RSD为2.83%、2.67%、1.90%、2.62%。不同产地4种成分的量存在显著性差异。通过聚类分析分为2类。结论:该方法简便、准确、分离效果好,所测为黄花菜中化学成分结果真实、可靠,可为其合理开发利用提供参考依据。
Objective: To determine four components from Hemerocallis citrina Baroni., such as quercetin, kaempferol, isorhamnetin and β-sitosterin by high performance liquid chromatography(HPLC), and to explore the scientific evidence for reasonable exploitation and utilization of medicinal Hemerocallis citrina Baroni.. Methods: Agilent C18 chromatographic column(250 mm×4.6 mm, 5μm) was used for detecting, 0.1% acetonitrile-phosphate water was taken as mobile phase, gradient elution was manipulated, volume flow rate of 1 ml/min and column temperature 30℃ were set, detection wavelength was 210 nm. SPASS software was used to analyze the measurement results. Results: Quercetin, kaempferol, isorhamnetin and β-sitosterin had the good linear relationship with peak area respectively in the range of 27.96-295.18, 27.78-304.64, 19.09-181.10, 3.86-55.43μg(r0.9992). The average recovery rate of four kinds of the ingredients was 99.25%, 99.06%, 98.80%, 96.91%, and RSD was 2.83%, 2.67%, 1.90%, 2.62%. Quantities of ingredients in four different regions had significant differences. By clustering analysis, it could be divided into two categories. Conclusion: This method is simple and accurate, and has good separate effect. The determined results of chemical composition in Hemerocallis citrina Baroni. are true and reliable, which can provide the reference for its rational development and utilization.
