摘要:目的:探讨当归与糖皮质激素及其合用对哮喘小鼠肺组织HMGB1的表达及阴虚症状的影响。方法:小鼠随机分为6组(n=12),采用卵蛋白致敏、激发来复制哮喘模型、利用氢化可的松来复制阴虚模型,观测受试药缓解阴虚症状(进食及饮水量、体重、自主活动)与平喘(哮喘行为学、肺功能、肺组织病理学)等作用,同时探讨当归与激素及其合用治疗哮喘的机制(细胞因子、HMGB1)。结果:8 g/kg当归与0.05 g/kg氢化可的松及其合用可明显缓解哮喘发作及其症状,提高肺功能,抑制炎性细胞浸润、改善肺组织病理学,降低BALF中TNF-α、IL-1β及IL-6水平,当归与氢化可的松合用后可抑制肺组织HMGB1的高表达。同时,当归与氢化可的松合用后可缓解糖皮质激素所致小鼠进食及饮水量增加、体重下降、自主活动增多等阴虚症状。结论:当归与激素及其合用具有明显的平喘作用,抑制HMGB1通路是当归与激素合用后的平喘机制之一;同时当归与激素合用后可缓解糖皮质激素引起的阴虚症状。
Objective: To study the effects of Radix Angelicae Sinensis(RASI),glucocorticoid and their combination on the expression of High-mobility group box 1(HMGB1) in lung tissue and the Yin deficiency symptom in the murine asthma model. Methods:The mice were randomly divided into 6 groups(n = 12). Used ovalbumin(OVA) to replicate asthma model and hydrocortisone sodium succinate(HSS) to copy Yin deficiency model,observed the effects of RASI,HSS and their combination on Yin deficiency symptoms(food and water intake,body weight and autonomic activities),asthma(asthmatic behaviors,lung function and histopathology),and antiasthma mechanisms(cytokines and HMGB1). Results: 8 g/kg RASI,0. 05 g/kg HSS and their combination could significantly relieve asthma attack and its symptoms,improve lung function,inhibit inflammatory cell infiltration,improve lung histopathology,and decrease the levels of TNF-α,IL-1β and IL-6 in broncho-alveolar lavage fluid(BALF). The combination of RASI and HSS could inhibit the high expression of HMGB1 in lung tissue. Moreover,RASI could alleviate yin deficiency symptoms induced by HSS in their combination,which include eating and drinking water increased,weight loss and increased autonomic activities. Conclusion: RASI,glucocorticoid and their combination have obvious anti-asthma effects and one of mechanisms is to down-regulate the expression of HMGB1 signaling pathway. RASI can alleviate yin deficiency symptom induced by glucocorticoid in the treatment of asthma.
