摘要:目的:基于生物信息学技术探讨白及多糖对胃溃疡的保护作用及机制,并进行实验验证。方法:利用GeneCards、OMIM等数据库对胃溃疡疾病相关靶点进行预测和筛选;通过String在线数据库进行蛋白互作分析;通过Cytoscape构建PPI网络,筛选核心靶点与白及多糖组分(葡萄糖、甘露糖)进行分子对接,60只大鼠随机分为空白组、模型组、阳性对照组和白及多糖高、低剂量组,各组大鼠分别给药干预7 d。除空白组外,其余大鼠全部通过盐酸/乙醇混合物复制胃溃疡模型。肉眼观察及病理检测评价大鼠胃组织损伤状况,并通过相关分子生物学技术对前期生物信息学技术预测得到的结果进行验证。结果:共筛选得到与胃溃疡密切相关的靶点56个,其中排名前3的是AKT、HIF-1α、HSP90,分子对接结果表明结合能皆<0。实验验证结果表明,与空白组比较,模型组大鼠胃组织溃疡性损伤显著,水肿充血坏死明显可见,镜下上皮细胞结构紊乱、坏死严重,病理学评分升高,AKT、HIF-1α、HSP90的基因蛋白表达水平均显著升高(P<0.01);与模型组比较,白及多糖显著降低溃疡指数(P<0.01),有效改善胃组织病理评分,显著降低AKT、HIF-1α的基因蛋白表达水平,升高HSP90基因蛋白表达水平(P<0.01,P<0.05)。结论:白及多糖具有显著的胃保护作用,其具体机制可能是葡萄糖和甘露糖能够显著调控胃组织凋亡、炎症、缺氧等病理状态,同时还能有效激活HSP90等保护性蛋白,进而发挥抗溃疡性损伤作用。
Objective:To explore the protective effect and mechanism of Bletilla Striata polysaccharide on gastric ulcer based on bioinformatics technology and experimental verification.Methods:Genecards,OMIM and other databases were used to predict and screen gastric ulcer disease-related targets;Protein protein interaction(PPI)analysis was carried out through String online database;The PPI network was constructed by Cytoscape,the core target was screened for molecular docking with Bletilla Striata Polysaccharide components(glucose and mannose).Sixty rats were randomly divided into blank group,model group,positive control group and Bletilla striata polysaccharide high-and low-dose groups.Rats in each group were treated for 7 d.Except for the blank group,all the other rats copied the gastric ulcer model through the hydrochloric acid/ethanol mixture.The gastric tissue was dissected,the gastric tissue damage was evaluated by naked eye observation and pathological detection,and the results predicted by previous bioinformatics technology were verified by relevant molecular biological technology.Results:The study showed that 56 targets closely related to gastric ulcer were screened,among which AKT and HIF-1αand HSP90 ranked the top three,molecular docking results show that the binding eneroy is less than 0.The experimental results showed that compared with the blank group,the ulcerative injury,edema,congestion and necrosis of gastric tissue in the model group were significant,the structure of epithelial cells was disordered and necrotic under microscope,the pathological score was increased,AKT,HIF-1αand HSP90 gene protein expression levels increased significantly(P<0.01).Compared with the model group,Bletilla Striata polysaccharide significantly reduced the gastric ulcer injury index(P<0.01),effectively improved the pathological score of gastric tissue,and significantly reduced the expression level of AKT,HIF-1αand HSP90 gene protein was increased(P<0.01,P<0.05).Conclusion:Bletilla Striata polysaccharide has significant gastric protective effect.The specific mechanism may be that glucose and mannose can significantly regulate the pathological states of gastric tissue such as apoptosis,inflammation and hypoxia.At the same time,it can effectively activate HSP90 and other protective proteins,so as to play an anti ulcer role.
