摘要:目的:采用网络药理学探究三黄凝胶治疗痤疮的作用机制。方法:通过传统中药系统药理学数据库与分析平台(TCMSP)查找三黄凝胶的化学成分和靶点;通过人类孟德尔遗传靶点数据库OMIM、治疗靶标数据库TTD、GeneCards和药物作用靶点数据库Drugbank检索痤疮疾病的靶点;运用UniProt对基因靶点标准化处理;应用Cytoscape软件构建药物成分-疾病-靶点的相互作用网络图;运用STRING平台搭建蛋白质-蛋白质相互作用网络;利用DAVID数据库预测靶点,进行基因本体论(GO)富集分析和京都基因与基因组百科全书(KEGG)通路富集分析。结果:三黄凝胶活性成分共有202个,预测出三黄凝胶治疗痤疮相关的靶点70个,预测出三黄凝胶通过作用于多条通路并介导多种生物学功能起到治疗痤疮的作用。蛋白质-蛋白质相互作用网络发现肿瘤坏死因子(Tumor Necrosis Factor,TNF)、白细胞介素(Interleukin,IL)-6、肿瘤蛋白p53(Tumor Protein p53,TP53)、血管内皮生长因子A(Vascular Endothelial Growth Factor A,VEGFA)、IL-1β、前列腺素内过氧化物合成酶2(Prostaglandin-endoperoxide Synthase 2,PTGS2)、JUN原癌基因(Jun Proto-Oncogene,JUN)、过氧化物酶体增殖物激活受体(Peroxisome Proliferator Activated Receptor Gamma,PPARG)、趋化因子蛋白8(Chemokine 8,CXCL8)、基质金属蛋白酶9(Matrix Metallopeptidase 9,MMP9)等可能是三黄凝胶治疗痤疮的关键靶点。GO功能富集分析得到生物过程条目115条,细胞组成条目6条,分子功能条目23条,主要涉及细胞因子介导的信号通路、转录的正向调控、基因表达的正向调控、炎症反应等生物过程,细胞表面、大分子复合物、细胞外区域等细胞组分,以及酶结合、锌离子结合、氧化还原酶的活性等分子功能。KEGG通路富集分析共富集到55条通路,主要与IL-17信号通路、丝裂原活化蛋白激酶信号通路、磷脂酰肌醇-3-激酶/蛋白激酶B信号通路、Th17细胞的分化、c型凝集素受体信号通路、缺氧诱导因子-1信号通路、雌激素信号通路、T细胞受体信号通路、核苷酸结合寡聚化结构域蛋白样受体信号通路等有关。结论:该研究首次揭示了三黄凝胶治疗痤疮具有多成分、多靶点、多通路协同作用的特点,为后续三黄凝胶防治痤疮作用机制的研究提供了参考依据。
bjective:To explore action mechanism of the Sanhuang gel(三黄凝胶)on acne by network pharmacology.Methods:The chemical components and targets of the Sanhuang gel were searched by TCMSP.The disease targets of acne were searched by OMIM,TTD,GeneCards and Drugbank.UniProt was used to standardize gene targets.Cytoscape software was used to construct the network interaction diagram of medicine components-disease-targets.A PPI network diagram was constructed by STRING platform.DAVID database was used to predict targets,and then the GO enrichment analysis and KEGG pathway enrichment analysis were performed.Results:There were 202 active components of the Sanhuang gel finally,and a total of 70 targets related to the treatment of acne were predicted,and the Sanhuang gel was predicted to play a role in the treatment of acne by acting on multiple pathways and mediating various biological functions.The PPI network showed that TNF,IL-6,TP53,VEGFA,IL-1β,PTGS2,JUN,PPARG,CXCL8 and MMP9 might be the key targets of the Sanhuang gel on acne.The GO functional enrichment analysis obtained 115 items of biological process,6 items of cell composition,and 23 items of molecular function,mainly involved cytokine-mediated signal pathway,positive regulation of transcription,positive regulation of gene expression,inflammatory reaction and other biological processes,cell components such as cell surface,macromolecular complex,and extracellular region,and molecular functions such as enzyme binding,zinc ion binding,and oxidoreductase activity.The KEGG pathway enrichment analysis has obtained 55 pathways,which were mainly related to IL-17 signaling pathway,MAPK signaling pathway,PI3K-Akt signaling pathway,Th17 cell differentiation,C-type lectin receptor signaling pathway,hypoxia inducible factor-1 signaling pathway,estrogen signaling pathway,T cell receptor signaling pathway,nod-like receptor signaling pathway,etc..Conclusion:This study reveals for the first time that the Sanhuang gel has the characteristics of multi-component,multi-target and multi-pathway synergistic effect in the treatment of acne.The result provides a reference for the following-up research on the action mechanism of the Sanhuang gel in preventing and treating acne.
