摘要:目的 通过生物信息学分析方法,系统评价miR-125a-5p在胃癌组织中的表达及其与临床病理特征的相关性;通过实验观察miR-125a-5p在胃癌组织中的表达,分析其临床意义,并采用生物信息学方法,进行靶基因和信号通路富集分析。方法 采用实时荧光定量聚合酶链式反应(PCR)法检测miR-125a-5p在胃癌组织和癌旁组织中的表达,并分析其临床意义;采用UALCAN数据库分析miR-125a-5p在胃癌组织中的表达及临床病理特征;采用miRecords数据库的预测软件预测其靶基因,选择至少3个以上软件支持的靶基因,运用DAVID 6.7在线富集分析靶基因参与的信号通路。结果 生物信息学分析和实验研究均发现,相对于癌旁组织,miR-125a-5p在胃癌组织中表达明显下调(P<0.05),其表达与肿瘤淋巴结转移、浸润深度、肿瘤组织增殖阶段、肿瘤组织类型相关(P<0.05);生物信息学分析提示,miR-125a-5p与临床病理特征关系密切,其靶基因富集在33个与肿瘤相关的信号通路中。结论 miR-125a-5p是胃癌中下调表达的分子标记物,是潜在的具有临床相关性的抑癌基因,是与临床病理特征关系密切的胃癌标记物。
Objective To evaluate systematically the expression of miR-125a-5p in gastric cancer tissues and the correlation with clinicopathological features by bioinformatics analysis, to experimentally observe the expression of miR-125a-5p in gastric cancer tissues and analyze its clinical significance, and to perform target gene and signaling pathway enrichment analysis by bioinformatics methods.Methods The expression of miR-125a-5p in gastric cancer tissues and paraneoplastic tissues was detected by real-time fluorescence quantitative polymerase chain reaction(PCR),and its clinical significance was analyzed.The expression and clinicopathological features of miR-125a-5p in gastric cancer tissues were analyzed using the UALCAN database.Its target genes were predicted using the prediction software of miRecords database, and at least three or more target genes supported by the software were selected, and the signalling pathways involved in the target genes were conducted online enrichment analysis using DAVID 6.7.Results Both bioinformatics analysis and experimental studies revealed that miR-125a-5p expression was significantly down regulated in gastric cancer tissues compared with paraneoplastic tissues(P<0.05),and its expression correlated with tumour lymph node metastasis, depth of infiltration, tumour tissue proliferation stage and tumour tissue type(P<0.05);Bioinformatics analysis suggested that miR-125a-5p was closely related to clinicopatho-logical features and its target genes were enriched in 33 tumor-related signaling pathways.Conclusion MiR-125a-5p is a down-regulated molecular marker in gastric cancer and a potentially clinically related tumor suppressor gene, which is gastric cancer biomarkers closely related to the clinicopathological characteristics.
