摘要:目的:基于"脾主运化"理论探讨运脾消积法对厌食症幼龄大鼠肠道微生态的影响。方法:以病因模拟法制备大鼠厌食症模型,观察记录大鼠摄食量和体重的变化;分别以模型组、阳性对照组(健胃消食片0.45g/kg)、小儿开胃增食5.7 g/kg、11.4 g/kg组干预模型大鼠,连续42 d,另设正常组。收集粪便,以Illumina Mi Seq为测序平台,对其16S rRNA V3区的肠道菌群可操作分类单元(OTUs)数量、alpha与beta多样性、丰富度指数、多样性指数以及差异菌门与菌属进行综合性分析与评价,并对菌群代谢功能进行预测。结果:小儿厌食症模型大鼠中明显失调的OUT数量、丰富度指数Chao1和ACE、菌落多样性、属水平的棒状杆菌属(Corynebacterium)、Odoribacter属、YRC22属、普雷沃菌属(Prevotella)、Jeotgalicoccus属、瘤胃球菌属(Ruminococcus)、rc4-4属以及具有外源化合物降解和代谢功能的菌群以及能量代谢的相关菌群、多糖生物合成功能和代谢功能的菌群经小儿开胃增食合剂干预后均向正常或保护方向恢复。结论:小儿厌食症模型大鼠肠道微生物菌落的多样性发生了明显改变。小儿开胃增食合剂可明显加强保护性菌属的有益改变,减弱有害性菌属的损伤改变,调整菌群的多样性,使小儿厌食症菌群多样性及其功能恢复正常。
Objective:To investigate the efficacy of Yunpixiaoji therapy on Intestinal Microecology of Anorexia model in juvenile rats based on the theory of"Pizhuyunhua".Methods:The anorexia model rat was established by etiological simulation method.The changes of food intake and body weight were recorded.Rats were divided into 5 groups and administered for 42 days:the control group,the model group,the positive control group(0.45 g/k Jianweixiaoshi pills)and pediatric appetizer groups(11.4 g/kg,5.7 g/kg).The feces were collected,and the Illumina Mi Seq was used as the sequencing platform to manipulate the intestinal flora of the 16S rRNA V3 region.The(OTUs)number,alpha and beta diversity,richness index,diversity index,and differential analysis of bacteria and bacteria were performed,and the metabolic function of the flora was predicted.Results:After the intervention,the number of OUT,the richness index Chao1 and ACE,the colony diversity,the genus Corynebacterium,Odoribacter,YRC22 genera,Prevotella,the genus Jeotgalicoccus,the genus Ruminococcus,the rc4-4 genus,the flora with the degradation and metabolism of exogenous compounds,the related flora of energy metabolism,the biosynthesis function of polysaccharides and the metabolic function of the flora through the pediatric appetite supplement were all recovered to normal or protective direction.Conclusions:The diversity of intestinal microflora in rats with anorexia has significantly changed.Pediatric appetizer and food supplement can significantly enhance the beneficial changes of the protective fungi,weaken the damage of the harmful bacteria,adjust the diversity of the flora,and make the diversity and function of the anorexia genus in juvenile rats return to normal.
