摘要:目的研究利咽止咳喷雾剂对急性咽炎模型大鼠的抗炎作用及可能的作用机制。方法取SPF级SD大鼠60只,每组10只,除空白组外,其余咽喉部喷雾25%氨水,每次2喷,每天上、下午各1次,连续3 d复制急性咽炎模型。将模型大鼠随机分为模型组,阳性组,利咽止咳喷雾剂给药组,空白组及模型组喷雾蒸馏水,给药组按1喷、2喷、3喷,阳性组2喷,bid,连续7 d。实验结束后麻醉采血并处理,取咽喉组织送病检,采用全自动生化分析仪测定血清中生化指标,采用酶联免疫法(ELISA)测定血清中IL^(-1)、IL-6、TNF-α、CRP。结果与空白组比较,模型组、中、高剂量组及阳性组中GLO含量降低(P<0.05,P<0.01);阳性组白细胞降低(P<0.01),模型组和阳性组中性粒细胞均升高(P<0.05,P<0.01),中剂量组CRP含量降低(P<0.05),IL-6、IL^(-1)含量升高(P<0.05),阳性组、高剂组TNF-α含量降低(P<0.01);与模型组比较,各剂量组CREA含量升高(P<0.01),TNF-α含量降低(P<0.05,P<0.01),ALT含量降低(P<0.01),高剂量组AST含量降低,红细胞升高(P<0.05),低药组和阳性组ALP含量降低(P<0.05),阳性组、高剂组白细胞均降低(P<0.05,P<0.01),低剂量组和中剂量组淋巴细胞均升高(P<0.05),阳性组、低剂量及中剂量组CRP含量降低(P<0.05),中剂量组IL-6、IL^(-1)含量升高(P<0.05)。结论利咽止咳喷雾剂对急性咽炎模型大鼠具有一定的抗炎作用。其作用机制可能是通过抑制TNF-α、CRP等炎症因子的表达起到抗炎作用。
OBJECTIVEE To study the anti-inflammatory effects and potential mechanisms of the Liyan Zhike Spray in a rat model of acute pharyngitis.METHODS A total of 60 SPF-grade SD rats were used,with 10 rats in each group,Except for the blank group,all rats were administered 25%ammonia spray on the throat,2 sprays per time,twice daily,for 3 consecutive days to induce an acute pharyngitis model.The model group and blank group were sprayed with distilled water.The treatment groups received 1,2,or 3 sprays of the Liyan Zhike spray,twice daily for 7 consecutive days.After the treatment,rats were anesthetized,and blood samples were collected.Throat tissue samples were examined histologically,and serum levels of inflammatory markers(IL^(-1),IL-6,TNF-α,CRP)were measured using enzyme-linked immunosorbent assay(ELISA).Biochemical parameters were analyzed using an automatic biochemical analyzer.RESULTS Compared to the blank group,the model group,as well as the low,medium,and high-dose treatment groups,showed a significant decrease in GLO levels(P<0.05,P<0.01).The positive control group showed a reduction in white blood cell count(P<0.01),while the model and positive control groups showed an increase in neutrophil count(P<0.05,P<0.01).The medium-dose group showed a significant decrease in CRP levels(P<0.05),with an increase in IL-6 and IL^(-1) levels(P<0.05).The positive control and high-dose groups showed a significant decrease in TNF-αlevels(P<0.01).Compared to the model group,the treatment groups showed an increase in CREA levels(P<0.01)and a decrease in TNF-α(P<0.05,P<0.01)and ALT levels(P<0.01).The high-dose group also showed a decrease in AST levels and an increase in red blood cells(P<0.05).The lowdose and positive control groups showed a decrease in ALP levels(P<0.05).The positive control and high-dose groups showed a significant reduction in white blood cell count(P<0.05,P<0.01),while the low and medium-dose groups showed an increase in lymphocyte count(P<0.05).Additionally,CRP levels were reduced in the positive control,low-dose,and medium-dose groups(P
