摘要:目的:观察芍药苷对溃疡性结肠炎(UC)大鼠白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、白细胞介素4(IL-4)的影响。方法:将实验动物分为4组,采用TNBS/乙醇灌肠法造模,光镜观察结肠组织形态并评分,采用ELISA法检测大鼠血清IL-1β、TNF-α、IL-4水平。结果:模型组大鼠结肠组织黏膜层可见溃疡形成,且有明显的炎症、充血存在,证实造模成功。模型组大鼠血清IL-1β、TNF-α水平均高于空白组(P<0.05),而血清IL-4水平则低于空白组(P<0.01);治疗后,芍药苷组血清IL-1β、TNF-α水平均较模型组降低(P<0.05、P<0.01),而血清IL-4水平则较模型组升高(P<0.01)。结论:芍药苷能够通过下调IL-1β、TNF-α和升高IL-4的表达,调节肠道异常免疫反应、抑制炎症反应,修复溃疡,进而治疗UC。
Objective : To observe the influence of the Paeoniflorin on Interleukin - 1β ( IL - 1 β ) , TNF - αt ( Tumor Necrosis Factor-α )and Interleukin-4 (IL-4)in rats with ulcerative colitis( UC ). Methods:The experimental animals were divided into four groups to prepare the rat models by means of the TNBS/ethanol enema, observing the colon organization form under the light microscope and then giving the score ; the levels of IL - 1β, TNF - αand IL - 4 in rat serum were detected by enzyme linked im-munosorbent assay(ELISA). Results:Colon mucosa of the model group showed ulceration ,and had obvious inflammation and con-gestion, which meant the modeling succeeded. The contents of IL - 1β and TNF - α in rats in the model group were transparently higher than those in the blank control group( P 〈 0. 05 ), and the content of IL - 4 was lower than it( P 〈 0. 01 ) ; after treatment, compared with the model group,the Paeoniflorin group's contents of IL - 1β and TNF -α were reduced(P 〈0.05 ,P 〈0. 01 ), and the content of IL- 4 was increased( P 〈 0. 01 ). Conclusion:The Paeoniflorin can reduce the expressions of IL-1 15 and TNF or,increase the expression of IL - 4, regulate immune abnormalities in intestines, inhibit inflammation, repair ulcers, and cure UC.
