摘要:目的研究生姜、干姜、炮姜对胃寒证模型大鼠的能量代谢,探讨温热药性本质。方法SD大鼠随机分为正常组、模型组(0℃冰水灌胃+10℃泡浴+0.3 mol·L^(-1)冰氢氧化钠刺激)和生姜、干姜、炮姜3个给药组(在模型组基础上分别灌胃给予8.1 g·kg^(-1)生姜、干姜、炮姜),连续给药7 d。用紫外分光光度法测定大鼠肝组织中琥珀酸脱氢酶(SDH)、谷氨酸脱氢酶(GDH)等能量代谢指标,用酶联免疫吸附法测定促肾上腺皮质激素(ACTH)、多巴胺β羟化酶(DβH)、促甲状腺激素(TSH)指标。结果正常组、模型组和干姜、炮姜给药组的Na^(+)-K^(+)-ATP酶含量分别为(0.36±0.06)、(0.29±0.02)、(0.35±0.03)和(0.38±0.09)U·mgprot-1,Ca^(2+)-Mg^(2+)-ATP酶含量分别为(1.07±0.32)、(0.41±0.24)、(0.94±0.27)和(0.87±0.15)U·mgprot-1,GDH含量分别为(5.12±0.80)、(3.07±0.92)、(4.37±1.12)和(4.56±1.38)U·mgprot-1。模型组的上述指标与正常组比较,差异均有统计学意义(P<0.05,P<0.01);干姜、炮姜给药组的上述指标与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。正常组、模型组和生姜、干姜、炮姜给药组的SDH含量分别为(59.69±5.12)、(15.59±7.95)、(24.02±6.74)、(34.08±13.81)和(51.44±11.42)U·mgprot-1,DβH含量分别为(61.96±2.87)、(36.47±1.18)、(45.75±3.07)、(48.38±3.07)和(57.83±1.44)pg·mL^(-1),ACTH含量为(23.51±2.07)、(68.48±5.06)、(61.19±2.54)、(44.51±5.42)和(39.88±5.42)ng·L^(-1),TSH分别为(12.69±1.02)、(5.40±0.40)、(6.90±1.10)、(7.84±1.10)和(11.21±0.51)mU·L^(-1)。模型组的上述指标与正常组比较,差异均有统计学意义(P<0.05,P<0.01);生姜、干姜、炮姜给药组的上述指标与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。结论生姜、干姜、炮姜均可改善寒证模型大鼠的能量代谢,炮姜的改善效果最为显著,温热效应最强,干姜次之,生姜较弱。
Objective To study the energy metabolism of fresh ginger,dried ginger and baked ginger in rats with stomach-cold syndrome,and to explore the nature of warming medicinal properties.Methods SD rats were randomly divided into 5 groups:Normal group,model group(gavage with 0℃ice water+10℃cool water bath+stimulation with 0.3 mol·L^(-1)ice sodium hydroxide),and 3 doses of SJ,GJ,and PJ group(8.1 g·kg^(-1)fresh ginger,dried ginger and baked ginger on the basis of the model group)were given intragastric administration continuously for 7 days.The energy metabolism indexes such as succinate dehydrogenase(SDH)and glutamate dehydrogenase(GDH)in rat liver tissue were determined by UV spectrophotometry,and the indexes of adrenocorticotropic hormone(ACTH),dopamineβ-hydroxylase(DβH)and thyroid stimulating hormone(TSH)were determined by enzyme-linked immunosorbent assay.Results The contents of Na^(+)-K^(+)-ATPase in normal group,model group,dried gingergroup and baked ginger group were(0.36±0.06),(0.29±0.02),(0.35±0.03),(0.38±0.09)U·mgprot-1;the contentsof Ca^(2+)-Mg^(2+)-ATPase in these 4 groups were(1.07±0.32),(0.41±0.24),(0.94±0.27),(0.87±0.15)U·mgprot-1;the GDH contents of these four groups were(5.12±0.80),(3.07±0.92),(4.37±1.12),(4.56±1.38)U·mgprot-1,respectively.There were statistically significant differences in the above indexes betweenthe model group and the normal group(P<0.05,P<0.01).Compared with the model group,there were statisticallysignificant differences between the two drug administration groups(P<0.05,P<0.01).The SDH contents of normalgroup,model group,SJ,GJ and PJ group were(59.69±5.12),(15.59±7.95),(24.02±6.74),(34.08±13.81),(51.44±11.42)U·mgprot-1;the DβH contents of these 5 groups were(61.96±2.87),(36.47±1.18),(45.75±3.07),(48.38±3.07),(57.83±1.44)pg·mL^(-1);the ACTH contents of these 5 groupswas(23.51±2.07),(68.48±5.06),(61.19±2.54),(44.51±5.42),(39.88±5.42)ng·L^(-1);the TSHcontents of these 5 groups were(12.69±1.02),(5.40±0.40),(6.90±1.10),(7.84±1.10),(11.21±0.51)mU·L^(-1),respectively.There were statistically significant differences in the above indexes between the model groupand the normal group(P<0.05,P<0.01).Compared with the model group,the differences were statisticallysignificant(P<0.05,P<0.01).Conclusion Fresh ginger,dried ginger and baked ginger can all improve theenergy metabolism of rats with cold syndrome,and baked ginger has the most significant improvement effect,and thewarm and hot effect is the strongest,followed by dried ginger and fresh ginger is weaker.
