摘要:【目的】比较甘肃产地与新疆产地乌拉尔甘草及其内生细菌抑菌活性的差异性.【方法】采用组织块及平板划线法,分别对甘肃和新疆产地乌拉尔甘草内生细菌分离纯化,用打孔法分别筛选具有抑制肺炎链球菌、金黄色葡萄球菌的有效菌株,进一步进行理化鉴定;用平板对峙法和二倍稀释法,以抑菌直径、最小抑菌浓度为指标,比较各有效菌株与宿主及各有效菌株之间的抑菌活性.【结果】甘肃酒泉产乌拉尔甘草共分离出内生细菌162株,与空白组比较,对肺炎链球菌有抑制作用的3株,对金黄色葡萄球菌有抑制作用的11株;与宿主比较,JQYB006、JQYB037、JQYB050、JQYB070对金黄色葡萄球菌的抑菌活性与其宿主相近,综合比较JQYB037活性显著,最小抑菌浓度为4.01×10~5CFU/mL.新疆甘草共分离出内生细菌83株,与空白组比较,对肺炎链球菌有抑制作用的3株,对金黄色葡萄球菌有抑制作用的6株;与宿主比较,XJYB023对肺炎链球菌的抑菌活性与其宿主相近,XJYB051、XJYB053对金黄色葡萄球菌的抑菌活性优于其宿主(P<0.05).宿主间比较,甘肃乌拉尔甘草对金黄色葡萄球菌的抑菌活性优于新疆甘草(P<0.05);有效菌株间比较,JQYB006、JQYB050、JQYB070对金黄色葡萄球菌的抑菌活性与XJYB051、XJYB053相近或相似.【结论】甘肃乌拉尔甘草对肺炎链球菌的抑菌活性优于新疆乌拉尔甘草,且甘肃乌拉尔甘草及其内生菌对金黄色葡萄球菌的抑菌活性均优于新疆乌拉尔甘草.
【Objective】 To compare the antibacterial activities of Glycyrrhiza uralensis and its endophytic bacteria in Gansu and Xinjiang.【Method】 Endophytic bacteria from Gansu and Xinjiang Ural licorice were isolated and purified by tissue block and plate streak method.The effective strains with inhibition of Streptococcus pneumoniae and Staphylococcus aureus were screened by perforation method,and physical and chemical identification was carried out.The antibacterial activity between each effective strain and the host and each effective strain was compared by the sputum method and the double dilution method,and the bacteriostatic diameter and the minimum inhibitory concentration were used as indicators.【Result】 A total of 162 endophytic bacteria were isolated from Gansu licorice.Compare with blank group,there were 3 strains inhibited pneumococcal bacteria and 11 strains inhibited S.aureus.Compared with licorice,the antibacterial activities of strains JQYB006,JQYB037,JQYB050,JQYB070 against S.aureus were similar to those of licorice.In comparison,the significant inhibitory concentration of JQYB037 was 4.01×10~5 CFU/mL.A total of 83 endophytic bacteria were isolated from Xinjiang licorice.Compare with blank group,there were 3 endophytic bacteria inhibiting pneumococcal bacteria and 6 strains inhibiting Staphylococcus aureus.Compared with licorice,the antibacterial activity of XJYB023 against S.pneumoniae was similar to that of its host.The antibacterial activity of XJYB051 and XJYB053 against S.aureus was better than that of its host(P<0.05).Comparing the hosts,the antibacterial activity of Gansu licorice against S.aureus and was better than that of Xinjiang licorice(P<0.05).Compared with the effective strains,the antibacterial activities of JQYB006,JQYB050 and JQYB070 against S.aureus were similar to those of XJYB051 and XJYB053.Compared with the effective strains,the antibacterial activities of JQYB006,JQYB050 and JQYB070 against S.aureus were similar or similar to those of XJYB051 and XJYB053.【Conclusion】 Gansu licorice has better antibacterial activity against S.pneumoniae and St.aureus than Xinjiang licorice.
