摘要:目的观察当归红芪超滤膜提取物对过氧化氢(H2O2)致PC12细胞氧化损伤的影响。方法 H2O2诱导PC12细胞构建氧化应激损伤模型;不同剂量当归红芪超滤膜提取物(0.38、0.75、1.5 g/L)干预;MTT法测定细胞存活率;生化法检测细胞内MDA、LDH、SOD含量;双氯荧光黄乙酸乙酯(DCF-DA)染色,检测细胞内ROS含量。结果 200μmol/L H2O2作用6 h成功诱导PC12细胞氧化应激损伤;与空白对照组比较,H2O2模型组细胞存活率下降,细胞内LDH、MDA、ROS含量增多,SOD含量下降(P<0.05);与H2O2模型组相比,当归红芪超滤膜提取物各剂量组细胞存活率明显上升,细胞内LDH、MDA、ROS含量降低,SOD含量增加(P<0.05)。结论当归红芪超滤膜提取物可修复H2O2诱导的PC12细胞氧化损伤。
Objective To observe the effects of ultrafiltration extract from angelica sinensis and hedysarum polybotrys on PC12 cells injury induced by hydrogen peroxide. Methods The oxidative damage cell model was established by H2O2- induced PC12 cells. The oxidative damaged cells were treated with three different dosages of ultra- filtration( 1. 5 g / L,0. 75 g / L,0. 375 g / L) extracted from angelica sinensis and hedysarum polybotrys respectively. The cell survival rate was measured by MTT method and the contents of MDA,LDH and MDA were detected by Ultraviolet spectrophotometry. The content of ROS was tested by DCF- DA staining. Results The oxidative damage of PC12 cells model was established by effect 6 hours in200 μmol / L H2O2 Compared with the blank control group,the survival rate of PC12 cells and the activity of SOD were decreased and the contents of LDH,MDA and ROS were increased in model group( P < 0. 05).Compared with the model group,the survival rate and the activity of SOD of PC12 cells were increased and the contents of LDH,MDA and ROS of PC12 cells were decreased in three different dosages of ultra- filtration ultrafiltration extract from angelica sinensis and hedysarum polybotrys( P < 0. 05). Conclusions The ultra- filtration extracted from angelica sinensis and hedysarum polybotrys have the repairable effect on H2O2- induced oxidative damage of PC12 cells.
