摘要:目的:探讨镇肝熄风汤对自发性高血压大鼠(SHR)胆囊收缩素(CCK) mRNA及其蛋白的影响。方法:14周龄雄性SHR 75只随机分为模型组、贝那普利0. 0009g/kg组、镇肝熄风汤15、30、60 g/kg组,以Wistar Kyoto大鼠(WKY)作为正常对照,模型组及正常对照组每日灌服同体积的蒸馏水,干预8周后,应用酶联免疫法检测血清中CCK的含量;采用RT-PCR法检测十二指肠中CCK mRNA表达;采用Western blot法检测十二指肠和脑中CCK蛋白表达。结果:干预8周后,与正常对照组比较,模型组十二指肠中CCK mRNA和蛋白表达、脑中CCK蛋白表达均显著降低,但血清CCK无变化;与模型组比较,贝那普利组、镇肝熄风汤15g/kg组十二指肠中CCK mRNA和蛋白表达显著升高,30g/kg组蛋白亦显著增高;各给药组脑中CCK蛋白表达显著升高;与贝那普利组比较,镇肝熄风汤15g/kg组十二指肠CCK mRNA及镇肝熄风汤60g/kg组蛋白表达显著升高,镇肝熄风汤30g/kg组脑中CCK蛋白表达显著升高。结论:SHR十二指肠和脑组织中CCK调节失衡,贝那普利和镇肝熄风汤的降压作用可能通过调节CCK的表达而达到降压目的,而镇肝熄风汤的调节作用更为明显。
Objective: To investigate the influence of Zhengan Xifeng decoction on the expressions of mRNA and protein of Cholecystokinin( CCK)in spontaneously hypertensive rats( SHR). Methods: 75 SHRs( 14 weeks old,male) were randomly assigned into the model group,0.0009g/kg/d Benazepril group and three doses of Zhengan Xifeng decoction groups( 15 g/kg、30 g/kg、60g/kg),and the normal control group( Wistar Kyoto rats). Rats in the model group and WKY group were given the same volume of distilled water. After 8 weeks’ administration,the content of CCK in serum was detected by enzyme linked immunoassay and the expression of CCK mRNA in duodenum was detected by RT-PCR. Also,the expressions of CCK protein were detected by Western blot in the duodenum and the brain. Results: Compared with WKY group,the expressions of CCK mRNA and protein in the duodenum and brain were significantly decreased,while the expressions of mRNA and protein of CCK in serum had no change. Compared with the model group,the expressions of CCK mRNA and protein in duodenum were significantly increased in Benazepril group and Zhengan Xifeng decoction groups( 15 g/kg、30 g/kg),and the expression of CCK protein in the brain increased in all groups. Compared with Benazepril group,the expressions of duodenal CCK mRNA and CCK protein in 15 g/kg and60 g/kg doses of Zhengan Xifeng decoction groups were significantly increased,and the expression of cerebral CCK protein was increased in30 g/kg dose of Zhengan Xifeng decoction group. Conclusions: The regulation disorder of CCK exists in the duodenum and the brain of SHR. The antihypertensive effect of Benazepril and Zhengan Xifeng decoction may be related to regulating the expression of CCK,while Zhengan Xifeng decoction shows better regulating effect.
