摘要:目的:探讨平痫冲剂抗癫痫的机制。方法:腹腔注射戊四唑制成慢性癫痫模型,通过免疫组织化学方法和显微图像分析技术检测大鼠颞叶及海马齿状回的谷氨酸受体(NMDAR1)和γ-氨基丁酸受体(GABA-ARα1)免疫反应的阳性细胞数(n)、阳性细胞面积比(Aa%)及阳性细胞积分吸光度(IA)。结果:(1)与正常对照组比较,模型组颞叶及齿状回NMDAR1的阳性细胞数、阳性细胞面积比上升,阳性细胞积分吸光度增加(P<0.01);而西药治疗组及中药小、中、大剂量组与模型组比较,颞叶及齿状回的NMDAR1阳性细胞数、阳性细胞面积比都有不同程度地下降,阳性细胞积分吸光度也有不同程度地减少(P<0.01)。(2)与正常对照组比较,模型组颞叶及齿状回GABA-ARα1阳性细胞数、阳性细胞面积比下降,阳性细胞积分吸光度降低(P<0.01);而西药治疗组和中药小、中、大剂量组与模型组比较,颞叶及齿状回的GABA-ARα1阳性细胞数、阳性细胞面积比都有不同程度地增加,阳性细胞积分吸光度也有不同程度地升高(P<0.01)。结论:平痫冲剂抗癫痫的作用机制可能是通过抑制颞叶及齿状回兴奋性神经递质受体NMDAR1的活性与表达,并激活其抑制性神经递质受体GABA-ARα1的活性与表达,从而降低大脑皮质的兴奋性,有效抑制癫痫的形成及发展。
Objective :To explore the possible antiepileptic mechanism of Pingxian Powder.Methods : Pentylenetetrazole (PTZ)was injected into abdomen of every rat to reconstruct epileptic kindling model. The total number,percentage of positive area and integral optical density of N-methyl-D-aspartate reeeptorl (NMDAR1) and γ-aminobutyrie acid-A reeeptor(GABA-ARα1) of immunoreactive cells in the hippocampus DG and the temporal cortex were studied by the immunohistochemieal method and image analyzing system. Results: The percentage of positive area and integral optical density of NMDAR1 of immunoreactive cells in the hippocampus DG and the temporal cortex of model group was more than control group(P 〈 0.01),while these numerical values of Western Medicine group and small,middle, high dose group of Pingxian Powder decreased to varying degrees constrasting with model group (P 〈 0.01). The percentage of positive area and integral optical density of GABA-ARα1 of immunoreactive cells in the hippocampus DG and the temporal cortex of model group was less than control group(P 〈 0.01 ),while these numerical values of Western Medicine group and small, middle, high dose group of Pingxian Powder enhanced to varying degrees eonstrasting with model group (P 〈0.01).Conclusion:Pingxian Powder may reduce the excitability of cerebral cortices by inhibiting the activities of NMDAR1 and promoting the activities of GABA-ARα1 in the hippocampus DG and the temporal cortex to inhibit the formation and development of epilepsy.
