摘要:目的观察党参多糖对大鼠溃疡性结肠炎的保护作用,并探讨其对结肠上皮NF-κB信号通路的影响。方法 50只雄性Wistar大鼠随机分为5组(每组10只),分别为正常组、模型组、柳氮磺胺嘧啶(SASP)阳性对照组(0.3 g/kg)、党参多糖高剂量组(9 g/kg)、党参多糖低剂量组(3 g/kg),采用三硝基苯磺酸(TNBS)/乙醇联合灌肠制作大鼠实验性溃疡性结肠炎(UC)模型。给药21 d后,观察大鼠疾病活动指数(DAI)、结肠黏膜损伤指数(CMDI)及结肠组织形态,生化法及ELISA法分别检测结肠组织中SOD(超氧化物歧化酶)活性、MDA(丙二醛)、白介素IL-6、IL-10和TNF-α(肿瘤坏死因子-α)水平,RT-PCR检测结肠组织中IL-6、IL-10、TNF-α mRNA表达,免疫组化法及Western blot法测定结肠组织中NF-κB蛋白表达。结果党参多糖高低剂量组均能降低UC大鼠DAI评分、CMDI评分,改善结肠黏膜形态,降低结肠组织中MDA、IL-6、TNF-α水平,提高结肠组织中SOD活性和IL-10水平(P<0.01),抑制IL-6、TNF-α mRNA表达,促进IL-10 mRNA表达(P<0.01),抑制NF-κB蛋白表达(P<0.01)。结论党参多糖具有保护大鼠溃疡性结肠炎的作用,其机制可能与抗脂质过氧化、抑制NF-κB信号通路减少炎症因子释放有关。
AIM To observe the protective effect of Codonopsis pilosula polysaccharides(CPPs)on rats’ulcerative colitis(UC),and to investigate their effect on colonic epithelial NF-κB signaling pathway.METHODS Fifty male Wistar rats were randomly divided into 5 groups(10 rats in each group),namely the normal group,model group,salazosulfadiazine(SASP)positive control group(0.3 g/kg),low-dose and high-dose CPPs groups(3,9 g/kg),with UC rat models established using trinitrobenzene sulfonic acid(TNBS)/ethanol and enema.After 21 days of consecutive corresponding administration,the rats were subjected to the observation of disease activity index(DAI),colonic mucosal injury index(CMDI)and colon tissue morphology.;the detection of colonic SOD activity,MDA,IL-6,IL-10 and TNF-αlevels by biochemical and ELISA methods;the detection of colonic IL-6,IL-10,TNF-αmRNA expression by RT-PCR;the determination of colonic protein NF-κB expression by histochemical and Western-Blot method.RESULTS CPPs groups shared reduced DAI score,CMDI score,and improved morphology of the colon mucosa in UC rats;reduced levels of MDA,IL-6,and TNF-α,and increased SOD activity and IL-10 levels in the colon tissue(P<0.01);inhibited IL-6 and TNF-αmRNA expression,and promoted IL-10 mRNA expression(P<0.01);and significantly decreased colonic NF-κB protein expression as well(P<0.01).CONCLUSION The protective effects of CPPs upon UC rats may contribute to their generated anti-lipid peroxidation,inhibition of NF-κB signaling pathway and inflammatory factors release.
