摘要:目的研究酶法-超声提取白花蛇舌草(Hedyotis diffusa)总黄酮的最佳工艺。方法以总黄酮含量为指标,通过单因素实验考查酶的种类、乙醇浓度、酶用量、酶解时间、料液比5个因素对白花蛇舌草总黄酮提取率的影响,并在此基础上通过中心组合实验设计(Central composite design,CCD)和响应面分析(Response surface methodology,RSM)优化白花蛇舌草总黄酮的提取工艺。结果当选用纤维素-果胶-木蛋白酶,乙醇浓度为60%、酶量为0.7%、酶解时间为90 min、料液比为1∶25(g·mL^(-1))时,白花蛇舌草总黄酮提取率最高,为1.95%。结论通过响应面法优选的白花蛇舌草总黄酮提取工艺合理可行,可作为提取白花蛇舌草总黄酮的一种新方法被采用。
OBJECTIVE To optimize the extraction process of total flavonoids from Hedyotis diffusa,which have good anti-tumor effect,and improve the yield by combining enzyme extraction method and ultrasonic extraction method.METHODS Based on the total flavonoids content,the effects of enzyme type,ethanol concentration,enzyme dosage,enzymatic hydrolysis time and solid-liquid ratio on the extraction rate of total flavonoids from Hedyotis diffusa were studied by single factor experiment.Central composite experimental design(CCD)and response surface methodology(RSM)were used to optimize the extraction process.RESULTS When cellulose-pectin-wood protease was used,the ethanol concentration was 60%,the enzyme amount was 0.7%,the enzymatic hydrolysis time was 90 min,and the ratio of material to liquid was 1∶25(g·mL^(-1)).The extraction rate of total flavonoids from Hedyotis diffusa was 1.95%.CONCLUSION The optimized extraction process of total flavonoids from Hedyotis diffusa by response surface methodology is reasonable and feasible,and the operation is simple.It can be used as a new method for extracting total flavonoids from Hedyotis diffusa.
