摘要:目的:探讨益气温阳活血利水法对慢性肺源性心脏病(CPHD)大鼠水通道蛋白4表达的影响及具体机制。方法:MCT法构建CPHD模型大鼠45只,按随机数字表法分为对照组(Con组)、CPHD模型组(CPHD组)、实验组(Exp组)3组,每组15只。Con组不予注射MCT与药物,与其他组无差异;CPHD组给予建模,建模成功后给予安慰剂灌胃;Exp组按成人体重与剂量折算公式,换算成大鼠剂量给予益气温阳活血利水法汤剂灌胃。Western blot与qRT-PCR检测样本AQP4等表达,qRT-PCR检测IL-1β、IL-6、TNF-α各组表达。结果:建模成功后,CPHD组大鼠PaCO2、HR显著高于Con组,而CPHD组PaO2、SaO2、LVSP、LVdp/dtmax显著低于Con组,差异具有统计学意义(P<0.05);另外,Exp组PaCO2、HR较CPHD组显著下降,而PaO2、SaO2、LVSP、LVdp/dtmax较CPHD组显著升高,差异均有统计学意义(P<0.05)。无论在mRNA与蛋白水平,CPHD组AQP4表达显著低于Con组;另外,Exp组AQP4表达较CPHD组显著升高,差异具有统计学意义(P<0.05)。CPHD组IL-1β、IL-6、TNF-α表达显著高于Con组;另外,Exp组IL-1β、IL-6、TNF-α表达较CPHD组显著下降,差异具有统计学意义(P<0.05)。CPHD组pERK1/2、pNF-κB表达显著高于Con组,而Exp组pERK1/2、pNF-κB表达较CPHD组显著下降,差异具有统计学意义(P<0.05)。结论:益气温阳活血利水法能通过上调心肌细胞AQP4表达,抑制炎症反应改善CHPD大鼠心脏功能与血液氧化情况,这可能是益气温阳活血利水法治疗CHPD的重要分子机制。
Objective:To investigate the effect of invigorating Qi,warming Yang,promotingblood circulation and inducing diuresis on aquaporin 4 expression in rats with chronic pulmonary heart disease(CPHD) and its mechanism.Methods: Forty-five CPHD rats were established by MCT method and randomly divided into three groups:control group(Con group),CPHD model group(CPHD group) and experimental group(Exp group),with 15 rats in each group.In the Con group,MCT and drugs were not injected,but there was no difference between the other groups.CPHD group was modeled and given placebo after successful modeling.Exp group was given invigorating Qi,warming Yang,promotingblood circulation and inducing diuresis for rats according to the formula of adult weight and dose conversion.Western blot and qRT-PCR were used to detect the expression of AQP4,and qRT-PCR was used to detect the expression of interleukin-1 beta(IL-1 beta),IL-6 and TNF-alpha.Results: After successful modeling,PaCO2 and HR in CPHD group were significantly higher than those in Con group,while PaO2,SaO2,LVSP and LVdp/dtmax in CPHD group were significantly lower than those in Con group(P<0.05).In addition,PaCO2 and HR in Exp group were significantly lower than those in CPHD group,while PaO2,SaO2,LVSP and LVdp/dtmax in Exp group were significantly higher than those in CPHD group.There was statistical significance(P<0.05).The expression of AQP4 in CPHD group was significantly lower than that in Con group,regardless of the level of protein and mRNA.In addition,the expression of AQP4 in Exp group was significantly higher than that in CPHD group(P<0.05).The expressions of IL-1 beta,IL-6 and TNF-alpha in CPHD group were significantly higher than those in Con group.In addition,the expressions of IL-1 beta,IL-6 and TNF-alpha in Exp group were significantly lower than those in CPHD group(P<0.05).The expressions of pERK1/2 and pNF-kappa B in CPHD group were significantly higher than those in Con group,while the expressions of pERK1/2 and pNF-kappa B in Exp group were significantly lower than those in CPHD group(P<0.05).Conclusion: Invigorating Qi,warming Yang,promotingblood circulation and inducing diuresis can improve cardiac function and blood oxidation in CHPD rats by up-regulating the expression of AQP4 in myocardial cells and inhibiting inflammation.
