摘要:目的探讨“调气明目”穴方对单眼弱视模型大鼠视皮层17区PKC/ERK1/2信号通路的影响。方法60只SPF级14天龄SD幼鼠随机分成5组。除空白组外其余各组复制弱视模型。阻断剂组和针刺+阻断剂组腹腔注射PD98059溶液。阻断剂1h后,针刺组和针刺+阻断剂组针刺“调气明目”穴方,疗程14d。P-VEP检测大鼠双眼视觉电生理变化;Western blot和ELISA测定PKC、ERK1/2、p-ERK1/2、p-CREB、c-Fos蛋白表达及含量。结果左眼P100波潜时和振幅各组差异均不显著(P>0.05);右眼模型组较空白组潜时延长、振幅降低(P<0.01),针刺组较模型组和针刺+阻断剂组均有潜时缩短、振幅升高(P<0.05);空白组和针刺组PKC、p-ERK1/2和p-CREB蛋白表达均高于模型组(P<0.01),针刺组显著高于针刺+阻断剂组(P<0.05),针刺+阻断剂组高于阻断剂组(P<0.05);阻断剂组PKC、p-ERK1/2和c-Fos蛋白与针刺+阻断剂组相比差异不显著(P>0.05);各组间ERK1/2表达差异不显著(P>0.05)。结论“调气明目”穴方可激活单眼剥夺弱视模型大鼠视皮层17区PKC/ERK1/2信号通路,改变突触可塑性,促进视功能恢复。
Objective To investigate the effect of the acupoint prescription of“Tiaoqi Mingmu”on the PKC/ERK1/2 signaling pathway in 17th area of visual cortex of rats with monocular amblyopia.Methods Sixty 14-day-old SD mice were randomly divided into five groups.Except for the blank group,the other groups replicated the amblyopia model.The blocker group and the acupuncture+blocker group were intraperitoneally injected with PD98059 solution.One hour after the blocker,the acupuncture group and the acupuncture+blocker group were given the“Tiaoqi Mingmu”acupoint prescription for a course of 14 days.P-VEP was used to detect the visual electrophysiological changes of the rat's binoculars;Western blot(WB)and ELISA were used to determine the protein expression and content of PKC,ERK1/2,p-ERK1/2,p-CREB,and c-Fos.Results There was no significant difference in the latency and amplitude of the P100 wave in the left eye among all groups(P>0.05).Compared with the blank group,the model group had prolonged latency and reduced amplitude in the right eye(P<0.01);the acupuncture group had shorter latency and increased amplitude compared with the model group and the acupuncture+blocker group(P<0.05);the protein expression of PKC,p-ERK1/2 and p-CREB in the blank group and acupuncture group were both higher than the model group(P<0.01),the acupuncture group was significantly higher than the acupuncture+blocker group(P<0.05),and the acupuncture+blocker group was higher than the blocker group(P<0.05).There was no significant difference between the blocker group and the acupuncture+blocker group in PKC,p-ERK1/2 and c-Fos proteins(P>0.05).There was also no significant difference in ERK1/2 expression between the groups(P>0.05).Conclusion“Tiaoqi Mingmu”acupoint prescription can activate the PKC/ERK1/2 signaling pathway in 17th area of the visual cortex of rats with monocular deprivation amblyopia model,change synaptic plasticity,and promote the recovery of visual function.
