摘要:目的:优化筛选大孔树脂富集纯化白花蛇舌草总黄酮的工艺条件,并测定其体内抗肿瘤的作用。方法:采用紫外分光光度法测定总黄酮含量,以吸附率、解析率为指标,对5种不同性能的树脂D-101、AB-8、NKA-9、HDP-826及聚酰胺进行筛选,结合单因素试验考察大孔树脂纯化总黄酮的工艺参数;建立MFC荷瘤小鼠模型,随机将其分为模型组、环磷酰胺(25 mg/kg)阳性对照组、白花蛇舌草总黄酮高、中、低剂量组(200、100、50 mg/kg),白花蛇舌草总黄酮灌胃给药,环磷酰胺腹腔注射,1次/d,给药周期8 d。给药结束后,计算抑瘤率及脏器指数,探讨总黄酮体内抑瘤作用。结果:最佳纯化工艺为D-101树脂,上样质量浓度为2.184 mg/mL,上样体积为80 mL,树脂的上样量为16 mL/g,上样体积流量为4 mL/min,水洗脱体积为50 mL,醇洗脱为80%乙醇,洗脱体积为50 mL,洗脱体积流量为4 mL/min,纯品占总黄酮粗提物的28.62%,纯度为57.5%。白花蛇舌草总黄酮对MFC荷瘤小鼠的肿瘤生长具有明显的抑制作用(P<0.05)。结论:该纯化方法稳定可靠,适用于纯化白花蛇舌草总黄酮,纯化后的白花蛇舌草总黄酮具有良好抗肿瘤活性。
Objective:To optimize the technological conditions for enrichment and purification of total flavonoids from Oldenlandia diffusa by macroporous resin,and to study its anti-tumor effect in vivo.Methods:The content of total flavonoids from Oldenlandia diffusa were detected by using ultraviolet spectrophotometry.Five kinds of resins D-101,AB-8,NKA-9,HDP-826 and polyamide with different properties were screend to determine the best resin suitable for purification of total flavonoids by taking the adsorption rate and resolution rate as the investigation indexes.Single factor experiment was combined to investigate the process parameters of purifying the total flavonoids by the best resin.The MFC tumor-bearing mice model was established and randomly divided into the model group,the Cyclophosphamide(25mg/kg)positive control group and Oldenlandia diffusa total flavonoids groups of high-dose(200 mg/kg),medium-dose(100 mg/kg)and low-dose(50 mg/kg).Oldenlandia diffusa total flavonoids were administered intragastrically and Cyclophosphamide was injected intraperitoneally for 8 days.After the treatment,the anti-tumor rate and organ indexes were calculated to explore the anti-tumor effect of the total flavonoids of Oldenlandia diffusa in vivo.Results:The best purification process was D-101 resin,the concentration of sample loading was 2.184 mg/mL,the volume of sample loading was 80mL,the volume of resin loading was 16 mL/g,the volume of sample loading was 4 mL/min,the elution volume was 50mL,the elution volume was 50mL by 80%ethanol,and the elution volume was 4 mL/min.Lastly,the pure product accounted for 28.62%in the total flavonoids crude extract,and the purity was 57.5%.The total flavonoids of Oldenlandia diffusa had obvious inhibitory effect on tumor growth of MFC tumor-bearing mice(P<0.05).Conclusion:The purification method is stable and reliable,which is suitable for the purification of the total flavonoids of Oldenlandia diffusa.The purified total flavonoids has a good anti-tumor activity.
