摘要:目的研究香砂六君子汤对慢性萎缩性胃炎(CAG)大鼠胃组织Bcl-2相关凋亡促进因子(BAD)、B淋巴细胞瘤-XL(Bcl-XL)、Bcl-2相关X蛋白(BAX)基因及其蛋白表达水平的影响。方法用综合法复制CAG动物模型。按随机数字表法将造模成功Wistar大鼠随机分为5组:模型组、对照组和高、中、低3个剂量实验组;另取正常大鼠作为正常组,每组均20只。正常组和模型组大鼠给予馏水灌胃,高、中、低3个剂量实验组分别给予香砂六君子汤24,12,6g·kg^-1·d^-1灌胃,对照组给予维酶素0.30 g·kg^-1·d^-1灌胃,持续给药120 d。用免疫印迹法检测BAD、Bcl-XL和BAX蛋白表达水平(灰度值),用实时荧光定量聚合酶链反应法检测BAD、Bcl-XL和BAX基因表达水平(荧光值)。结果正常组、模型组、对照组和高、中、低3个剂量实验组的BAD蛋白表达量分别为0.24±0.06,1.23±0.28,0.58±0.07,0.41±0.13,0.80±0.10和1.07±0.20;这6组的Bcl-XL蛋白表达量分别为0.42±0.02,2.52±0.29,0.84±0.09,0.47±0.02,0.85±0.16和2.01±0.26;这6组的BAX蛋白表达量分别为1.17±0.37,0.36±0.05,0.51±0.07,1.07±0.21,0.59±0.09和0.44±0.05。上述指标:模型组与正常组比较,差异均有统计学意义(均P<0.01);高剂量实验组与模型组比较,差异均有统计学意义(均P<0.01)。基因表达结果与蛋白表达的趋势一致。结论香砂六君子汤通过上调胃组织BAX基因及其蛋白表达,下调BAD、Bcl-XL基因及其蛋白表达,达到影响CAG的作用。
Objective To study the effects of Xiangsha Liujunzi Decoction on Bcl-2-related apoptotic promoting factor(BAD),B lymphoma-XL(Bcl-XL)and Bcl-2-related X protein(BAX)gene and its protein expression in gastric tissue of rats with chronic atrophic gastritis(CAG).Methods The CAG model rats were replicated by comprehensive methods.Wistar rats successfully modeled were randomly divided into five groups by random number table:model group,control group and experimental-H,experimental-M,experimental-L groups;another normal rats as normal group,each group had 20 rats.Normal group and model group were given distilled water;experimental-H,experimentalM and experimental-L groups were respectively given 24,12,6g·kg^-1·d^-1of Xiangsha Liujunzi Decoction;control group was given0.30 g·kg^-1·d^-1of vitacoenzyme,for 120 d.The protein expressions(grey value)of BAD,Bcl-XL and BAX in gastric tissues of rats were detected by Western blot.The gene expressions(threshold fluorescence)of BAD,Bcl-XL and BAX in gastric tissues of rats were detected by real-time fluorescence quantitative polymerase chain reaction.Results The expression levels of BAD protein in gastric tissues of rats in normal group,model group,control group,experimental-H,experimental-M and experimental-L group were 0.24±0.06,1.23±0.28,0.58±0.07,0.41±0.13,0.80±0.10 and1.07±0.20;the expression levels of Bcl-XL protein in the six groups were 0.42±0.02,2.52±0.29,0.84±0.09,0.47±0.02,0.85±0.16 and 2.01±0.26;the expression levels of BAX protein in the six groups were1.17±0.37,0.36±0.05,0.51±0.07,1.07±0.21,0.59±0.09 and 0.44±0.05.The above three indicators showed that the differences between model group and normal group were statistically significant(all P<0.01);the differences of the indicators between experimental-H and model group were statistically significant(all P<0.01).The trend of gene results is consistent with that of protein.Conclusion Xiangsha Liujunzi Decoction can up-regulate the expressions of BAX gene and its proteins in gastric tissue,down-regulate the expressions of BAD and Bcl-XL genes and its proteins,and then affect the effect of CAG.
