摘要:目的:研究归芪聪志汤对血管性痴呆(varscular dementia,VD)模型大鼠学习记忆和海马神经元紧密连接蛋白1(Claudin-1)和转化生长因子-β(transforming growth factor-β,TGF-β)表达的影响。方法:Wistar雄性大鼠120只,按随机数字表选10只大鼠作为假手术组,其余大鼠制备VD模型,选成功模型大鼠随机分为5组,分别为模型组、阳性组和归芪聪志汤低、中、高剂量组。归芪聪志汤低、中、高剂量组给药量分别为9.9,19.8,39.6 g·kg-1·d-1。阳性组给予脑复康吡拉西坦胶囊0.3 g·kg-1·d-1。假手术组、模型组给予等量水。每日1次。连续灌胃治疗28 d后,Morris水迷宫予以行为学检测,实时荧光定量PCR(Real-time PCR),免疫组织化学(immunohistochemistry,IHC)与蛋白免疫印迹法(Western blot)检测各组海马组织Claudin-1,TGF-β基因和蛋白表达。结果:灌胃治疗28 d后,与假手术组比较,VD模型组大鼠的平均逃避潜伏期明显延长、跨原平台次数明显减少(P〈0.01),大鼠海马的Claudin-1 mRNA及蛋白表达水平明显升高(P〈0.05),大鼠海马的TGF-βmRNA及蛋白表达水平明显降低(P〈0.05);与模型组比较,各治疗组大鼠逃逸潜伏期均缩短,跨原平台次数均增多(P〈0.01),大鼠海马的Claudin-1 mRNA及蛋白表达水平明显降低(P〈0.05),大鼠海马的TGF-βmRNA及蛋白表达水平明显升高(P〈0.05),归芪聪志汤中剂量组与脑复康吡拉西坦胶囊组优于其他组(P〈0.05)。结论:归芪聪志汤能够显著改善VD大鼠的学习记忆能力,可能与调节脑组织细胞间黏附作用、抑制炎性因子表达等有关。
Objective: To observe the effect of Guiqi Congzhi decoction on expressions of Claudin-1,transforming growth factor-β( TGF-β) in the hippocampus area of vascular dementia( VD) rats. Method: Ten rats were selected from 120 male wistar rats as sham-operation group according to random number table. The remaining rats were selected to prepare the VD model; then,the successfully modeled rats were randomly divided into model group,positive control group,and high,medium and low-dose Guiqi Congzhi decoction groups( 9. 9,19. 8,39 g·kg-1·d-1). The positive group was given Piracetam capsules( 0. 3 g·kg-1·d-1). The sham operation group and the model group were given the equal amount of water,respectively. The medicines were administered once a day. All of the groups were intervened with the corresponding medicines through gastric perfusion once a day after modeling,which lasted for 28 days. Then,rats' behaviors were detected by Morris water maze,the protein expressions of Claudin-1 and TGF-β in hippocampus of rats were detected by IHC and Western blot,the mRNA expressions of Claudin-1 and TGF-β in hippocampus were detected by Real-time PCR. Result: Compared with sham-operation group,the average escaping latency period was significantly prolonged,and the number of crossplatform was decreased distinctly in VD rats( P〈0. 01),the mRNA and protein expressions of Claudin-1 were apparently increased in hippocampus of VD rats( P〈0. 05),the mRNA and protein expressions of TGF-β was apparently declined in hippocampus of VD rats( P〈0. 05); compared with VD model group,the average escaping latency period was shorted obviously,and the number of cross-platform was increased distinctly in middle-dose Guiqi Congzhi decoction group and positive control group( P〈0. 01),the mRNA and protein expressions of Claudin-1 were apparently increased in hippocampus of middle-dose Guiqi Congzhi decoction group( P〈0. 05),the mRNA and protein expressions of TGF-β was apparently increased in hippocampus of middle-dose Guiqi Congzhi decoction group and positive control group( P〈0. 05). Conclusion: Guiqi Congzhi decoction could improve the learning and memory abilities of VD rats. The mechanism may be correlated with the regulation of the adhesive effect between brain tissues and cells and the inhibition of the expressions of inflammatory factors.
