摘要:目的研究红芪多糖对实验性脾虚大鼠胃黏膜中肿瘤坏死因子-α(TNF-α)表达的影响。方法将60只Wistar大鼠随机分为6组,即空白对照组(A组)、模型对照组(B组)、黄芪多糖高剂量对照组(C组)、黄芪多糖中剂量对照组(D组)、红芪多糖高剂量组(E组)、红芪多糖中剂量组(F组),每组10只,其中A组大鼠不予特殊处理,正常饲养,其余5组采用苦寒泻下法制备脾虚证模型。造模成功后,C,D,E,F组分别灌胃高、中剂量的黄芪多糖或红芪多糖溶液30 d,灌胃后处死大鼠,取胃标本进行免疫组化染色,光镜下观察,计算大鼠胃黏膜中TNF-α的阳性表达率。结果B组TNF-α阳性表达率与A组比较显著升高(P<0.01);与B组比较,C,D,E,F各组TNF-α阳性表达率显著降低,差异有高度统计意义(P<0.01);C,D组之间,E,F组之间的TNF-α阳性表达率差异均有高度统计意义(P<0.01),高剂量组均低于中剂量组。结论红芪多糖能够降低脾虚大鼠胃黏膜中TNF-α的表达,能提高胃黏膜的修复能力,从而防止胃黏膜发生进一步病变。
Objective To study the effects of Hedysaryum polysaccharide on the expression of TNF-α in gastric mucosa of experimental rats with spleen deficiency.Methods 60 Wistar rats were randomly divided into 6 groups: blank control group(A group),model control group(B group),high and middle dose control group of astragalus polysaccharides(C and D group),high and middle dose group of Hedysaryum polysaccharide(E and F group),10 rats per group.The rats of A group weren't given special handle,were fed normally.The others rats were made the models of syndrome of spleen deficiency by adopting the purging method.After making model,the rats of C,D,E and F group had been administrated with high and middle dose of astragalus polysaccharides and Hedysaryum polysaccharide solution for 30 days.The rats were put to death,the specimen of stomach were taken out and given immunohistochemical staining,observed under optical mirror,and calculated the positive expression rate of TNF-α in gastric mucosa of rats.Results The positive expression rate of TNF-α in B group was significantly higher than A group(P0.01).Compared with B group,the positive expression rates of TNF-α in C,D,E and F group were significantly reduced(P0.01).There was high statistical significance between C and D,E and F(P0.01),high dose groups were lower than middle dose groups.Conclusion Hedysaryum polysaccharide can reduce the expression of TNF-α in gastric mucosa of rats with spleen deficiency,improve the repair capacity of gastric mucosa,thus prevent that further lesions of gastric mucosa is occurred.
