摘要:目的:观察痛泻要方对肝郁脾虚型溃疡性结肠炎(ulcerative colitis,UC)模型大鼠Casepase3、Chop、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的影响,分析痛泻要方对UC的疗效及可能的作用机制。方法:将60只SPF级Wistar大鼠随机分为空白组,模型组,痛泻要方高剂量组、中剂量组、低剂量组,美沙拉嗪组,每组各10只。各组给予相应药物,21d后取材。采用ELISA法检测大鼠血清TNF-α水平,采用SP和实时荧光定量PCR法检测结肠组织Casepase3、Chop蛋白和基因水平。结果:与模型组比较,美沙拉嗪组、痛泻要方高剂量组TNF-α水平降低(P <0. 05);与模型组比较,美沙拉嗪组,痛泻要方高剂量组、中剂量组Casepase3、Chop蛋白的表达减小,Casepase3、Chop mRNA表达减小(P <0. 01或P <0. 05)。结论:痛泻要方对肝郁脾虚型UC大鼠模型血清TNF-α表达、结肠黏膜Chop、Casepase3基因和蛋白的表达有下调作用,提示痛泻要方治疗UC的作用可能与调节炎症细胞因子、抑制内质网应激诱导的细胞凋亡有关。
Objective: To observe the effects of Tongxie Yaofang on Casepase 3,Chop and tumor necrosis factor-α( TNF-α) in rats with ulcerative colitis( UC) due to stagnation of liver qi and spleen deficiency,and to analyze the curative effect and possible mechanism of Tongxie Yaofang on UC. Methods: 60 SPF Wistar rats were randomly divided into blank group,model group,Tongxie Yaofang high-dose group,middle-dose group,low-dose group and mesalazine group with 10 rats in each group. The corresponding drugs were given to each group,and the samples were taken 21 days later. Serum TNF-α levels were detected by ELISA,and Casepase 3,Chop protein and gene levels were detected by SP and real-time fluorescence quantitative PCR. Results: Compared with model group,the levels of TNF-α in Mesalazine group and Tongxie Yaofang high-dose group decreased( P < 0. 05);compared with model group,the expressions of Casepase 3 and Chop protein in Tongxie Yaofang high-dose group and middle-dose group decreased,while the expressions of Casepase 3 and Chop mRNA decreased( P < 0. 01 or P < 0. 05). Conclusion: Tongxieyaofang can down-regulate the expression of TNF-α in serum,Chop,Casepase3 gene and protein in colon mucosa of UC rats due to stagnation of liver qi and spleen deficiency,suggesting that Tongxie Yaofang ’s effect on UC may be related to regulating inflammatory cytokines and inhibiting endoplasmic reticulum stress-induced apoptosis.
