摘要:目的 研究炙红芪和炙黄芪对脾气虚大鼠脑肠功能的影响。方法 取SPF级雄性大鼠50只,其中10只作为空白组,40只采用过度疲劳、限制饮食、苦寒泻下三因素复合造模。模型复制成功后随机分为模型组、炙红芪组、炙黄芪组和补中益气丸组,炙红芪和炙黄芪组灌胃1.26 g·mL^(-1)水煎液,补中益气丸组灌胃1.89 g·mL^(-1)水煎液,给药量10 mL·kg^(-1),空白组和模型组灌胃同体积蒸馏水,实验中测定大鼠体质量、肛温及游泳时间。给药结束,测定大鼠胸腺及脾指数,用酶联免疫吸附(ELISA)法测定大鼠血清白细胞介素-6(IL-6)、干扰素-γ(IFN-γ)、肿瘤坏死因子α(TNF-α)、环氧合酶-2(COX-2)、二胺氧化酶(DAO)、分泌型免疫球蛋白A(SIgA)、多巴胺(DA)及血管活性肠肽(VIP)含量。结果 模型组较空白组大鼠体质量、肛温及游泳时间均显著降低,脾及胸腺指数显著降低;炙红芪组和炙黄芪组能够改善脾气虚大鼠的胸腺质量、胸腺指数、脾质量和脾指数。空白组、模型组、炙红芪组、炙黄芪组和补中益气组大鼠血清IL-6含量分别为(63.88±1.75)、(98.82±6.62)、(80.77±7.57)、(80.42±3.93)和(70.23±2.86)pg·mL^(-1),IFN-γ含量分别为(1 302.14±115.90)、(400.71±62.21)、(671.43±65.68)、(625.71±96.58)和(960.00±39.05)pg·mL^(-1),TNF-α含量分别为(119.05±4.96)、(287.76±26.78)、(188.10±13.16)、(210.98±16.41)和(140.74±7.21)pg·mL^(-1),COX-2含量分别为(17.22±4.77)、(46.08±5.52)、(27.30±4.50)、(27.86±3.56)和(21.40±3.68)pg·mL^(-1),DAO含量分别为(89.49±5.81)、(158.06±8.39)、(111.65±6.70)、(120.48±5.43)和(109.97±6.91)pg·mL^(-1),SIgA含量分别为(79.01±4.99)、(42.01±4.34)、(68.54±6.33)、(59.54±7.55)和(72.72±8.33)μg·mL^(-1),DA含量分别为(78.31±9.85)、(48.09±6.66)、(71.45±11.05)、(64.33±4.38)和(67.59±12.52)pg·mL^(-1),VIP含量分别为(59.56±6.67)、(142.64±8.10)、(108.35±6.61)、(106.85±15.78)和(74.29±4.02)pg·mL^(-1)。模型组的上述指标与空白组比较,炙红芪组、炙黄芪组的上述指标与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。结论 炙红芪和炙黄芪可能通过调节免疫、脑和胃肠道功能达到对脾气虚的治疗效果。
Objective To study the effects of honey-processed Hedysari Radix and honey-processed Astragalus on brain and intestinal function in rats with spleen qi deficiency.Method Fifty SPF grade male rats were selected,of which 10 were used as blank group,and 40 were used as a composite model of excessive fatigue,dietary restriction,and bitter cold diarrhea.After successful replication of the model,the rats were randomly divided into model group,honey-processed Hedysari Radix group,honey-processed Astragalus group,and Buzhong Yiqi Pills group.Honey-processed Hedysari Radix and Honey-processed Astragalus groups were given 1.26 g·mL^(-1)decoction by gavage,while the Buzhong Yiqi Pills group was given 1.89 g·mL^(-1)decoction by gavage,with a dosage of 10 mL·kg^(-1).The blank group and model group were given the same volume of distlled water by gavage.The body weight,anal temperature,and swimming time of the rats were measured in the experiment.At the end of administration,the thymus and spleen indices of rats were measured.Rat serum was taken and enzyme-linked immunosorbent assay(ELISA)was used to measure the levels of interleukin-6(IL-6)and interferon-(IFN-γ),tumor necrosis factor-α(TNF-α),cyclooxygenase-2(COX-2),diamine oxidase(DAO),secretory immunoglobulin A(SIgA),dopamine(DA),and vasoactive intestinal peptide(VIP).Results The model group had lower body weight,anal temperature,and swimming time compared to the blank group;the spleen and thymus index were significantly reduced;and the treatment groups of honey-processed Hedysari Radix and honey-processed Astragalus could improve the thymus mass,thymus index,spleen mass,and spleen index of spleen deficiency rats.The serum IL-6 levels of rats in blank group,model group,honey-processed Hedysari Radix group,honey-processed Astragalus group and Buzhong Yiqi Pills group were(63.88±1.75),(98.82±6.62),(80.77±7.57),(80.42±3.93)and(70.23±2.86)pg·mL^(-1);the IFN-γ contents were(1302.14±115.90),(400.71±62.21),(671.43±65.68),(625.71±96.58)and(960.00±39.05)pg·mL-^(1);the TNF-αcontents were(119.05±4.96),(287.76±26.78),(188.10±13.16),(210.98±16.41)and(140.74±7.21)pg·mL^(-1);the COX-2 contents were(17.22±4.77),(46.08±5.52),(27.30±4.50),(27.86±3.56)and(21.40±3.68)pg·mL^(-1);the DA0 contents were(89.49±5.81),(158.06±8.39),(111.65±6.70),(120.48±5.43)and(109.97±6.91)pg·mL^(-1);the SIgA contents were(79.01±4.99),(42.01±4.34)(68.54±6.33),(59.54±7.55)and(72.72±8.33)μg·mL^(-1);DA content were(78.31±9.85),(48.09±6.66),(71.45±11.05),(64.33±4.38)and(67.59±12.52)pg·mL^(-1),respectively;VIP contents were(59.56±6.67),(142.64±8.10),(108.35±6.61),(106.85±15.78)and(74.29±4.02)pg·mL^(-1),respectively.Model group compared with blank group,honey-processed Hedysari Radix and honey-processed Astragalus groups compared with model group,the difference were all statistically significant(P<0.05,P<0.01).Conclusion Honey-processed Hedysari Radix and honey-processed Astragalus may achieve therapeutic effects on spleen qi deficiency by regulating immune,brain,and gastrointestinal functions.
