摘要:目的:观察痛泻要方治疗溃疡性结肠炎(ulcerative colitis,UC)的疗效,并探讨其作用机制。方法:采用TNBS/乙醇灌肠法制备结肠炎大鼠模型,光镜观察结肠组织病理形态变化并评分,采用ELISA法检测大鼠血清IL-2、IL-10、IL-6水平。结果:与空白对照组比较,模型组大鼠血清IL-2、IL-6含量均明显升高,IL-10含量显著下降,差异具有统计学意义(P<0.01);痛泻要方高、中、低剂量组血清IL-2、IL-6水平均较模型组降低,而IL-10水平明显升高,差异具有统计学意义(P<0.05)。结论:痛泻要方能够通过下调促炎因子IL-2、IL-6的表达,升高抗炎因子IL-10的表达,促进抗炎和抑炎因子的平衡,调节肠道异常免疫反应、抑制炎症反应,修复溃疡,进而防治UC。
Objective:To observe the curative effect of Tongxie prescription on ulcerative colitis(UC)and to investigate the mechanism.Methods: To prepare the rat models by means of the TNBS/ethanol enema,observe the colon tissue pathological morphology under the light microscope,then give it the score;and to detect the levels of IL-2,IL-10 and IL-6 in rat serums.Results:Compared with the blank control group,the content of IL-2,IL-6 in rats in the model group was transparently high,and the content of IL-10 in rat in the model group was decreased significantly,and the discrepancy was statistically significant(P0.01);the content of IL-2 and IL-6 in rats in the prescription low,moderate-high-dose groups were lower than that in the model group,and the content of IL-10 was transparently high,and the discrepancy was statistically significant(P0.05).Conclusion:Tongxie prescription can decrease the expression of lL-2 and IL-6,increase the expression of IL-10,promote the balance between the anti-inflammatory and inhibit inflammation factor,regulate immune abnormalities in intestines,inhibit inflammation,repair ulcers and cure UC.
