摘要:骨质疏松症(osteoporosis,OP)是一种以骨量减少、骨微结构破坏为显著特征的骨代谢性疾病,是当今困扰中老年人群最普遍的骨骼疾病之一。然而在OP的防治方面,至今仍未发现有效且安全的靶向药物。中药作为传统医学体系中的重要组成部分,在OP的防治上有着丰富的实践经验和深远的潜力。红景天苷(salidroside,SAL)是红景天属植物中广泛存在的一种天然酚类次生代谢产物,近年来SAL治疗骨质疏松的研究越来越多。具体表现出多方面的作用机制,包括通过PI3K/Akt、HIF-1α/VEGF、OPG/RANKL/RANK、BMP2、Wnt/β-catenin及TGF-β/Smad2/3等多条信号途径直接调控成骨、破骨细胞增殖及分化,还可发挥抗氧化应激、调节脂代谢、维生素代谢等作用,维持体内外骨代谢平衡,改善OP。本文回顾了当前国内外SAL对骨组织代谢影响的相关文献,对其具体分子机制进行了总结,旨在强调其在预防和治疗骨质疏松症中的潜在作用,以期为防治OP进一步研究提供参考依据。
Osteoporosis(OP)is a bone metabolic disease significantly characterized by decreased bone mass and destruction of bone microstructure,and is one of the most prevalent musculoskeletal diseases afflicting theelderly population today.However,no effective and safe targeted drugs have been found for the prevention and treatment of OP.As an important part of traditional medicine system,Chinese medicine has rich practical experience and far-reaching potential in the prevention and treatment of OP.Salidroside(SAL)is a natural phenolic secondary metabolite widely found in Rhodiola rosea plants,and in recent years SAL has been increasingly studied for the treatment of osteoporosis.Specifically,it exhibits multifaceted molecular mechanisms,including the direct regulation of osteoblast and osteoclast proliferation and differentiation through multiple signaling pathways,such as PI3K/Akt,OPG/RANKL/RANK,HIF-1α/VEGF,BMP2,etc.It also exerts the roles of anti-oxidative stress,regulation of lipid metabolism,etc.,to maintain bone metabolic homeostasis and improve the osteoporosis symptomatology.In this paper,we review the current literature related to the effects of SAL on bone tissue metabolism at home and abroad,and summarize its specific molecular mechanisms,aiming to emphasize its potential role in the preventionand treatment of osteoporosis,with a view to providing a reference basis for further research on prevention and treatment of OP.
