摘要:目的探讨葛根芩连汤保护2型糖尿病大鼠胰岛细胞的可能作用机制。方法 24只雄性ZDF(fa/fa)大鼠给予高脂饲料诱导2型糖尿病模型,造模成功后随机分为葛根芩连汤组、二甲双胍组、模型组,每组8只。8只雄性ZDF(fa/+)大鼠设为正常组。模型组、正常组给予生理盐水10 ml/kg灌胃,葛根芩连汤组给予葛根芩连汤13.8g/kg灌胃,二甲双胍组给予盐酸二甲双胍肠溶片300 mg/kg灌胃。给药8周后检测空腹血糖(FPG),采用蛋白免疫印迹法检测胰腺胰岛素受体底物2(IRS-2)、磷脂酰肌醇3激酶(PI3K)p85、蛋白激酶B(Akt2)、磷酸化胰岛素受体底物2(p-IRS2)、磷酸化磷脂酰肌醇3激酶(pPI3K)p85、磷酸化蛋白激酶B(p-Akt2)的蛋白表达。实时荧光定量PCR法检测胰腺IRS-2、PI3K p85与Akt2 mRNA表达。ELISA法测定空腹胰岛素(FINS)含量,免疫抑制比浊法测定糖化血红蛋白(Hb A1c)的百分含量。结果与模型组比较,葛根芩连汤组FPG、FINS水平及Hb A1c下降,IRS-2、PI3K p85、Akt2、p-IRS2、p-PI3K p85、p-Akt2蛋白表达上升,IRS-2、PI3K p85和Akt2 mRNA表达上升(P<0.05或P<0.01)。结论葛根芩连汤可能是通过提高IRS-2/PI3K-Akt信号转导通路的活性起到保护胰岛β细胞的作用。
Objective To explore the possible mechanism of Gegen Qinlian Decoction (葛根芩连汤) in protecting islet cells of newly diagnosed type 2 diabetic rats. Methods A total of 24 male ZDF (fa/fa) rats were given high-fat diet to induce type 2 diabetes mellitus. After the model was successfully established, the rats were randomly divided into C, egen Qinlian Decoction group, metformin group, model group, with 8 rats in each group. 8 male ZDF (fa/+) rats were used as negative control group. The model group and negative control group were given by gavage with normal saline 10ml/kg, and Gegen Qinlian Decoction group was given by gavage with Gegen Qinlian Decoction 13. 8 g/kg. Mefformin group was given by gavage with metformin 300 mg/kg. After 8 weeks of administration, the expression of IRS-2, PI3K p85, Akt2, p-IRS2, p-PI3K p85 and p-Akt2 in the pancreas was detected by Western blot. The expres-sion of IRS-2, PI3K p85 and Akt2 mRNA was detected by RT-qPCR. The content of FINS was determined by ELISA. The percentage of HbAlc was determined by TINIA. Results Compared with model group, the FPG level, FINS levels and HbAlc ratio was declined, and IRS-2, PI3K, Akt protein expression, mRNA expression (P 〈0. 05 or P 〈 0. 01) and p-IRS2, p-PI3K p85, p-Akt2 protein expression (P 〈 0. 01) was increased in Gegen Qinlian Decoction group. Conclusion Gegen Qinlian Decoction may be through the enhancement of IRS-2/PI3K-Akt path-way to achieve the role of protection of pancreatic β-cells.
