摘要:目的:运用数据挖掘和网络药理学的方法分析中药治疗肥胖病的用药规律,并探讨其作用机制。方法:搜集中文期刊数据库中治疗肥胖的方剂,通过关联规则、聚类及复杂网络分析,并筛选出核心药物,运用网络药理学分析其治疗肥胖的机制。结果:共筛选出92篇文献,涉及99首方剂,154味中药,得到高频药物36味。用量分析显示,常用药物的用量范围较大,其中茯苓、白术、泽泻、甘草最常用剂量分别为15 g、15 g、10 g、6 g。GO富集分析结果显示,交互基因涉及的生物学过程为参与对药物的应答、细胞增殖的正调控、对缺氧的应答等。KEGG通路富集结果主要为TNF信号通路、HIF-1信号通路,其中与脂质代谢直接相关的为脂肪细胞因子信号通路。结论:健脾利湿、清热化痰是中药治疗肥胖的重要治疗原则。茯苓、白术等9药核心组方是治疗脾虚湿阻的基本组方,可通过多通路发挥作用,临床用药时可作为肥胖病的靶药的使用。
Objective:To explor the medication rule and action mechanism of traditional Chinese medicine in the treatment of obesity by data mining and network pharmacology.Methods:The prescriptions for the treatment of obesity in the Chinese journal database were collected, and association rules, clustering and complex network analysis were analyzed, and core drugs were obtained, and the mechanism of core prescription were analyzed by network pharmacology. Results:A total of 92 literatures were selected, involving 99 prescriptions and 154 drugs. 36 high-frequency drugs and their common doses were obtained. Dosage analysis showed that the dosage range of common drugs was large, among which the most commonly used doses of Fuling, Baizhu, Zexie, Gancao were 15 g, 15 g, 10 g and 6 g respectively. Go enrichment analysis showed that the interacting genes involved in biological processes such as drug response, positive regulation of cell proliferation, response to Hypoxia and so on. KEGG pathway was mainly involved in TNF pathway, HIF-1 pathway, and the APLN pathway was directly related to Lipid metabolism. Conclusion:Invigorating the spleen and removing dampness, clearing away heat and dissolving phlegm is the important therapeutic principle of Traditional Chinese Medicine in the treatment of obesity. the core prescription composed of the 9 core drugs is the basic prescription for the treatment of spleen deficiency and dampness resistance, which works through multiple channels. In clinical, it should be paid to the use of target drugs.
