摘要:目的探讨复方湿生扁蕾胶囊组方药物湿生扁蕾与苦豆子不同配伍比例时湿生扁蕾3种化学成分(木犀草素、当药黄素和当药醇苷)的变化及临床的合理应用。方法采用Agilent SB-C18色谱柱(250 mm×4.6 mm,5μm);甲醇和0.04%磷酸水溶液为流动相,在0~20 min内甲醇∶0.04%磷酸水溶液由40∶60~63∶37线性梯度洗脱,在20~50 min内甲醇∶0.04%磷酸水溶液(63∶37)等度洗脱,流速0.8 m L/min,检测波长254 nm,柱温30℃。结果木犀草素在30 min内达到基线分离,回归方程为Y=84 800X+4 540(r=0.999 9),线性范围为0.166~1.664μg;当药黄素在15 min内达到基线分离,回归方程为Y=52 300X-32 300(r=0.999 9),线性范围为0.169~4.240μg;当药醇苷在20 min内达到基线分离,回归方程为Y=102 000X+83 400(r=0.999 9),线性范围为0.179~1.792μg。当湿生扁蕾与苦豆子配伍比例为2 0∶0.9时,符合临床应用安全、有效的原则。结论复方湿生扁蕾胶囊以湿生扁蕾∶苦豆子=20∶0.9的配伍比例符合临床应用安全、有效的原则,为配伍的合理性提供了实验依据。
Objective To probe the changes and clinical reasonable application of the chemical compositions( luteolin,swertisin and swertianolin) when the Gentianopsis paludosa and Sophora alopecuroides are in different compatible proportions in the Compound Shishengbianlei Capsule. Methods The samples were separated on the column of Agilent SB-C18( 250 mm × 4. 6 mm,5 μm) which moving phase was methanol and 0. 04% H3PO4 water solution. Linear grading elution was used at 40∶ 60 ~ 63 ∶ 37( methanol∶ 0. 04% H3PO4water) in 0 ~ 20 min;isocratic elution was used at 63∶ 37( methanol∶ 0. 04% H3PO4water) in 20 ~ 50 min. The detected wave-length was at 254 nm,flow rate was 0. 8 m L / min,column temperature was 30 ℃. Results Luteolin baseline separation was achieved within 30 minutes,the regression equation was Y = 84 800 X + 4 540( r = 0. 999 9),the linear range was 0. 166 ~ 1. 664 μg; swertisin baseline separation was achieved within 15 min,the regression equation was Y =5 2 300X- 32 300( r = 0. 999 9),the linear range was 0. 169 ~ 4. 240 μg; swertianolin baseline separation was a chieved within 20 min,the regression equation was Y = 102 000 X + 83 400( r = 0. 999 9),the linear range was 0. 179 ~ 1. 792 μg. When the compatible proportion of Gentianopsis paludosa to Sophora alopecuroides was 20 to 0. 9,it was safe and effective in clinical application. Conclusion It's safe and effective in clinical application when the compatible proportion is 20∶ 0. 9. It also provides the experimental basis for the rationality of the compa- tibility.
