摘要:目的 通过多指标正交试验优选白蕾颗粒最佳提取及纯化工艺。方法 以芍药苷、甘草苷、当药黄素和木犀草素的提取率及出膏率为指标,优选白蕾颗粒最佳提取工艺;以芍药苷、甘草苷、当药黄素和木犀草素的转移率及出膏率为指标,优选白蕾颗粒最佳纯化工艺;通过重复试验对优选工艺进行验证。结果 最佳提取工艺为:加10倍量水,提取30min,共提取3次;最佳纯化工艺为:浓缩至相对密度1.06~1.08(60℃),加95%乙醇至药液含醇量60%,室温下静置12h;优选工艺验证试验RSD值均小于5.98%。结论 本试验优选的提取纯化工艺稳定可靠,重复性好,可为该制剂后续开发提供科学依据。
Objective To optimize the extraction and precipitation process of Bailei Granules by multi index orthogonal experiment. Methods With extraction rate of paeoniflorin, glycyrrhizin, swertisin, luteolin and yield rate of dry extract as indexes, the extraction process of Bailei Granules was optimized. Taking the transfer rate of paeoniflorin, glycyrrhizin, swertisin, luteolin as indexes, the alcohol precipitation process of Bailei Granules was optimized, and optimized technology was verified by repeated tests. Results The optimal extraction process was as follows: add 10 times of water, extracting for 30min, 3 times;The optimal purification process was: concentrated to the relative density of 1.06~1.08(60℃),95% ethanol was added to make the alcohol content up to 60%,stand at room temperature for 12h;The RSD values of the validation tests of the optimized process were all less than 5.98%.Conclusion The optimized extraction and purification process was stable, reliable and reproducible, which could provide scientific basis for the subsequent development of the preparation.
