摘要:目的:探讨菖蒲郁金汤对多发性抽动症(TS)模型大鼠行为及纹状体单胺类神经递质的影响。方法:将雄性SD大鼠82只随机分为空白组12只,造模组70只,采用亚氨基二丙腈(IDPN)腹腔注射法制备TS动物模型,造模成功后,除去体质量偏差较大的10只大鼠,将其余大鼠分为模型组、泰必利组及中药高、中、低剂量组各12只。从造模后第2天开始灌胃给药,空白组与模型组给予纯净水,其余各组给予相应药物,1次/d,连续给药4周。观察各组大鼠行为评分变化及纹状体神经递质含量变化情况。结果:1)与模型组比较,泰必利组大鼠刻板行为及运动行为评分降低(P<0.05);中药高剂量组大鼠刻板行为及运动行为评分降低(P<0.05);中药中剂量组大鼠刻板行为评分降低(P<0.05)。2)与空白组比较,模型组多巴胺、去甲肾上腺素、5-羟色胺及高香草酸含量均无明显变化(P>0.05);与模型组比较,泰必利组多巴胺、去甲肾上腺素含量升高(P<0.01),中药高剂量组多巴胺、高香草酸含量升高(P<0.01),中药中剂量组多巴胺、高香草酸含量升高(P<0.05)。结论:菖蒲郁金汤具有降低模型大鼠刻板行为及运动行为评分的作用;具有抑制多巴胺受体超敏感、促进多巴胺代谢的作用,且其疗效与剂量有关。
Objective: To observe the effects of ChangPu YuJin decoction on the behavior and monoamine neurotransmitter in corpus striatum of Tourette syndrome (TS) rat model. Methods: All 82 male SD rats were randomized into 12 rats in the blank group and 70 rats in the model group, TS animal models were prepared by using intraperitoneal injection of IDPN, after successfully setting up the models, ten rats showing great body mass deviation were removed, the other rats were divided into the model group, Tiapride group, high, moderate and low dosages groups of herbs, 12 rats each group. The rats began to lavage since the second day after establishing the models, the blank group and the model group were given purified water, and the others were given corresponding herbs, once per day, for four weeks consecutively. The contents of striatal neurotransmitter and behavior scores of the rats in different groups were observed. Results: 1) Compared with the model group, the scores of stereotyped behavior and motor behavior of the rats in Tiapride group decreased (P〈0.05); the scores of stereotyped behavior and motor behavior of the rats in high dosage group of herbs lowered (P〈0.05); the scores of stereotyped behavior of the rats in moderate dosage group of herbs lowered (P〈0.05). 2) Compared with the blank group, the contents of dopamine, noradrenaline, 5-HOT and homovanillic acid in the model group showed no obvious changes (P〉0.05);compared with the model group, the contents of dopamine and noradrenaline raised (P〈0.01); the contents of dopamine and homovanillic acid of the rats in high dosage group of herbs raised (P〈0.01); the contents of dopamine and homovanillic acid of the rats in moderate dosage group of herbs evaluated (P〈0.05). Conclusion:ChangPu YuJin decoction could decrease the scores of stereotyped behavior and motor behavior of the rat model, inhibit hypersensitivity of dopamine receptor and promote the metabolism of dopamine, furthermore, its effects are related to the dosage.
