摘要:目的:观察归脾汤对苯中毒小鼠外周血、骨髓有核细胞计数及骨髓细胞凋亡蛋白Fas、Fas L表达的影响,初步揭示归脾汤可能减轻苯中毒对小鼠骨髓造血功能损害并提高外周血象的机理。方法:将72只小鼠随机分为正常对照组、模型组、西药组以及归脾汤大、中、小剂量组,每组12只。除正常对照组外,其余组进行熏苯复制苯中毒模型。造模后,归脾丸大、中、小剂量组分别给予归脾汤溶液8、4、2mg/(kg·d)灌胃;西药组给予利血生1.5mg/(kg·d)和鲨肝醇5mg/(kg·d)混悬液灌胃;正常对照组和模型组给予生理盐水灌胃,均每天1次,连续3周。应用全自动血细胞分析仪、流式细胞仪及SABC检测试剂,分别检测外周血细胞、骨髓有核细胞计数及Fas、Fas L蛋白的表达。结果:归脾丸可明显升高外小鼠周血白细胞(WBC)、血红蛋白(HGB)(P<0.05或P<0.01),而对血小板作用差别无统计学意义(P>0.05);与模型组比较,归脾汤高、低剂量组及西药组骨髓有核细胞计数均明显增加,差异有统计学意义(P<0.05);与模型组相比,归脾汤大剂量组、西药组能明显降低骨细胞凋亡蛋白Fas、Fas L的表达(P<0.05);与西药组相比,归脾汤大剂量组及小剂量组能对Fas、Fas L的影响无统计学意义(P>0.05)。结论:归脾汤可改善苯中毒小鼠骨髓造血组织,提高骨髓有核细胞计数以促进骨髓增生,同时也可降低骨细胞凋亡蛋白的表达,达到减轻苯中毒对小鼠骨髓造血功能的损害,改善外周血象。
Objective:To observe the effect of the Guipi decoction(归脾汤)on the peripheral blood,bone marrow nucleated cells and apoptosis protein Fas,FasL expression in mice with benzene poisoning and to reveal its possible mechanism in relieving the injure of bone marrow haematopoietic function in mice with benzene poisoning and increasing the levels of peripheral hemogram.Methods:Kunming mice(n=72)were randomly divided into the normal group,the model group and the western medicine group and high-dose,mid-dose and low-dose Guipi decoction groups(n=12).The benzolism model was established by putting mice into a manual simulated condition.The high-dose,mid-dose and low-dose groups were orally given solution of the Guipi decoction(0.072g/ml,0.036g/ml and0.018g/ml)respectively,and the normal group and the model group were orally given normal saline water(20ml/kg),once a day,for3weeks.The quantity of peripheral blood cell were detected by using automotive blood cell analyzer and flow cytometry;The nucleated bone marrow cell count were detected by using flow cytometry;and the expression of Fas and FasL protein were detected respectively by using SABC reagent.Results:The Guipi decoction could significantly improve the blood cell(WBC)and hemoglobin(HGB)(P<0.05or P<0.01),but it had no effect on the platelet(P>0.05);compared with the model group,the nucleated bone marrow cell count increased significantly in the high and low dose GPD groups,and the Western medicine treatment group(P<0.05);compared with the model group,the expression of Fas and FasL,bone marrow apoptosis proteins was decreased significantly(P<0.05);Compared with the western drug group,the changes of expression of Fas and FasL in high-dose group and Western medicine group was of no statistically significant difference(P>0.05).Conclusion:GPP could promote the hyperplasia of bone marrow through improving bone marrow haematopoietic tissue and increasing the count of nucleated bone marrow cell;meanwhile,GPP also relieved the injure of bone marrow haematopoietic function in mice with benzene poisoning and increased the levels of peripheral hemogram through decreasing the expression of bone marrow apoptosis proteins.
