摘要:目的:分析山茱萸炮制前后血清中移行成分的差异,从血清药物化学的角度揭示山茱萸炮制机理。方法:将山茱萸与制山茱萸水提液动物口服给药后制备含药血清,建立并分析比较空白组、山茱萸组和制山茱萸组大鼠血清HPLC特征图谱,确定山茱萸炮制前后血清移行成分差异,并进行主成分分析(PCA)和偏最小二乘判别分析(PLS-DA)分析。结果:与空白组相比,山茱萸组新增6个色谱峰,制山茱萸组新增5个色谱峰,两给药组共有色谱峰为11号、12号、13号色谱峰,其中11号和12号色谱峰峰面积存在显著性差异;S-1、S-2和S-3号色谱峰为山茱萸组特有,Z-1和Z-2号色谱峰为制山茱萸组特有。PCA和PLS-DA分析结果显示,山茱萸和制山茱萸组均出现明显的聚类,且两组间分离明显。结论:山茱萸炮制前后血清移行成分存在量与质的显著差异。
OBJECTIVE To analyze the difference of migrating components in serum before and after processing of Cornus officinalis and explore the processing mechanism of Cornus officinalis from the perspective of serum medicinal chemistry.METHODS The medicated serum was prepared by oral administration of Cornus officinalis and the aqueous extract of Cornus officinalis.The HPLC characteristic spectra of serum of rats in blank group and Cornus officinalis group were established and analyzed.The differences of serum migration components before and after processing of Cornus officinalis were determined.Principal component analysis(PCA)and partial least squares discriminant analysis(PLS-DA)were performed.RESULTS Compared with the blank group,6 chromatographic peaks were added in the Cornus officinalis group,and 5 chromatographic peaks were added in the processed Cornus officinalis group.The two common chromatographic peaks were 11,12 and 13 chromatographic peaks,of which11 and12 chromatographic peaks had significant differences in peak area;S-1,S-2 and S-3 chromatographic peaks were unique to the Cornus officinalis group,and Z-1 and Z-2 chromatographic peaks were unique to the Cornus officinalis group.The results of PCA and PLS-DA analysis showed that there was a significant clustering between Cornus officinalis and Cornus officinalis,and the separation between the two groups was obvious.CONCLUSION There are significant differences in the amount and quality of serum migration components before and after processing of Cornus officinalis.
