摘要:目的:优选铁棒锤种子中乌头碱的最佳提取方法。方法:分别采用浸泡法、超声提取法和回流法提取铁棒锤种子中乌头碱,采用HPLC方法测定其含量,色谱条件:ZORBAX Edipse XDB-C18色谱柱(4.6 mm×250 mm,5μm);G131513二极管阵列检测器;柱温30℃;流动相为乙腈-0.3%二乙胺水溶液(50∶50);流速1.0 m L/min;检测波长240 nm。比较分析测定结果,优选出最佳提取方法。结果:测得以浸泡法提取乌头碱的含量最高,为0.15%;超声提取法次之,为0.10%;回流法提取效果较差,为0.039%。结论:优选出铁棒锤种子中乌头碱的最佳提取方法为浸泡提取法,简便易行,重现性好。
Objective: To optimize the extraction technology for aconitine in the seed of Aconitum pendulum Busch. Methods:Aconitine was extracted from the seed of Aconitum pendulum Busch by immersion extraction, ultrasonic extraction and reflux method, respectively. Then the content of aconitine was determined by HPLC, and the extraction technology was optimized through the comparison and analysis of the results. Results: The content of aconitine was up to 0.15% by immersion extraction,then was ultrasonic extraction of 0.10%, and reflux method was the lowest of 0.039%. Conclusion: The best method was immersion extraction for extracting aconitine from the seed of Aconitum pendulum Busch.
