摘要:目的:探索家兔类风湿关节炎(RA)寒证模型的建立方法。方法:90只清洁级青紫蓝家兔随机分为空白组、卵蛋白组、卵蛋白+冷冻组,每组30只。卵蛋白组采用卵蛋白溶液与等量弗氏完全佐剂混成乳化剂,在家兔肩背部6个点各注射0.2m L,14d后再注射;卵蛋白+冷冻组注射相同乳化剂,在末次注射24h后冷冻1.5h,复温5min。冷冻后1d随机分为3亚组,观察各亚组冷冻后1、7、14d一般表现、膝周径、关节表面温度及关节滑膜病理积分变化。结果:空白组未见异常;与前者比较,卵蛋白+冷冻组、卵蛋白组膝关节肿胀、滑膜病理积分均增高(P<0.05);与同组冷冻后1d比较,冷冻后7d肿胀达到高峰、滑膜病理积分较高(P<0.05);与同组冷冻后7d比较,冷冻后14d膝关节肿胀减轻、滑膜病理积分减小(P<0.05),但仍较冷冻后1d积分高(P<0.05)。结论:采用卵蛋白诱导联合低温冷冻法建立的RA寒证模型可靠,模型至少可持续14d。
Objective: To explore the method of establishing cold syndrome animal model of rheumatoid arthritis(RA) in rabbits. Methods: A total of 90 healthy purple blue rabbits were randomly allocated into blank group, ovalbumin group, ovalbumin and freezing group, with 30 rabbits in each group. In ovalbumin group, animals were sensitized with ovalbumin Freund's complete adjuvant administered intradermally at six sites in the upper back. Two weeks later, a second sensitizing(booster) dose was administered; ovalbumin and freezing group was injected with the same emulsifier. Freezing for 1.5h after the last injection of 24 h, and then rewarming for 5min. After being frozen for 1 day, rabbits were randomly divided into 3 sub-groups. The changes of the general performance, the knee circumference, the local skin temperature and the joint synovial pathology scores were observed in each sub-group after being frozen for 1 day, 7 days and 14 days. Results: There was no abnormal changes in the blank group; compared with the former, the swelling of the knee joint and the synovial pathology scores in the ovalbumin group, the ovalbumin and freezing group were increased significantly(P<0.05); after 7 days, compared with the 1st day, the swelling of the knee-joints reached the peak and the synovial pathological scores were higher(P<0.05); 14 days later, compared with the 7th day, the swelling of the knee-joints and the synovial pathological scores were decreased significantly(P<0.05); however, they were still higher than the 1st day(P<0.05). Conclusion: The cold syndrome RA model established on the basis of ovalbumin-induced and freezing was reliable. The animal model lasted for at least 14 days.
