摘要:目的研究当归补血汤不同提取物HPLC指纹图谱及总多糖、总黄酮、总皂苷和总多酚与体外抗氧化活性之间的关系,明确发挥抗氧化活性的药效物质基础,筛选出最好的提取方法。方法建立当归补血汤不同提取物指纹图谱,测定4类化合物的含量;采用DPPH法、ABTS法和FRAP法测定不同提取物抗氧化活性;以含量和抗氧化药效为指标评价提取方法;利用灰色关联法分析当归补血汤不同提取物共有峰及4类化合物与综合抗氧化活性之间的相关性。结果建立了当归补血汤不同提取物HPLC指纹图谱,其中主要成分与4类化合物对抗氧化作用的大小顺序为总多酚>洋川芎内酯H>阿魏酸>洋川芎内酯I>总黄酮>总皂苷>芒柄花苷>芒柄花素>毛蕊异黄酮>未知成分1>总多糖>未知成分11>未知成分5>毛蕊异黄酮葡萄糖苷>绿原酸。结论基于灰色关联分析方法明确当归补血汤不同提取物抗氧化的药效是多种化学成分共同作用的结果,筛选出70%醇作溶剂回流提取方法最好。
Objective To investigate the HPLC fingerprints of different extracts of Danggui Buxue Decoction(当归补血汤,DGBXD)and the relationship between total polysaccharides,total flavonoids,total saponins and total polyphenols and antioxidant activity in vitro,to clarify the pharmacodynamic material basis for antioxidant activity,and to screen out the best extraction method.Methods Fingerprints of different extracts of DCBXD were established,and the contents of four types of compounds were determined.The antioxidant activities of different extracts were determined by DPPH,ABTS and FRAP.The content and antioxidant efficacy were used as indicators to evaluate the extraction method.The grey relational analysis(CRA)was used to analyze the correlation between the common peaks of different extracts of DGBXD,the four types of compounds and the comprehensive antioxidant activity.Results HPLC fingerprints of different extracts of DGBXD were established,and the order of antioxidant effect of the main components and four types of compounds was total polyphenols>senkyunolide H>ferulic acid>senkyunolide I>total flavonoids>total saponins>manganese>manganese ananthin>mullein isoflavones>unknown component 1>total polysaccharides>unknown component 11>unknown component 5>verbasin isoflavone glucoside>chlorogenic acid.Conclusion Based on GRA,it is clear that the antioxidant efficacy of different extracts of DGBXD is the result of the joint action of multiple chemical components,and the best method for solvent reflux extraction is to screen out 70%alcohol.
