摘要:目的研究丹参酮对去卵巢大鼠体质量、肝脏脂代谢的影响及其分子机制。方法将40只健康雌性SD大鼠随机分为假手术组、去卵巢组、雌激素组和丹参酮组,每组10只,实验进行50 d。记录大鼠的体质量和摄食量,检测大鼠的血糖、肝脏的甘油三脂和胆固醇的含量,以及成脂基因的表达,对所得结果进行分析。结果去卵巢组大鼠在体质量、血糖水平、肝脏甘油三脂、总胆固醇、成脂基因的表达,以及信号传导和转录激活因子3(signal transducer and activator of transcription 3,STAT3)的磷酸化水平方面都高于假手术组、雌激素组和丹参酮组(P<0.05)。结论丹参酮可以抑制去卵巢大鼠体质量的增加,丹参酮通过抑制去卵巢导致的STAT3磷酸化,抑制成脂基因的表达,降低肝脏甘油三酯和总胆固醇的含量,改善去卵巢导致的肝脏脂代谢异常。
Objective To study the effects of tanshinone on body weight and liver lipid metabolism in ovariectomized rats and its molecular mechanism.Methods Forty healthy female Sprague-Dawley rats were randomly divided into sham operation group,ovariectomy group,estrogen group and tanshinone group,10 rats in each group,and the experiment was carried out for 50 days.The body weight and food intake of the rats were recorded,and the blood glucose,liver triglyceride and cholesterol levels,and the expression of adipogenic genes were measured,and the results were analyzed.Results In the ovariectomized group,the body weight,blood glucose level,liver triglyceride,total cholesterol,adipogenic gene expression,and the level of phosphorylation of signaling and transcriptional activator 3(STAT3)were higher than in the SHAM group,estrogen group and tanshinone group(P<0.05).Conclusion Tanshinone inhibits weight gain in ovariectomized rats.By inhibiting the phosphorylation of STAT3 caused by OVX,tanshinone inhibits the expression of lipogenic genes,reduces the contents of triglyceride and total cholesterol in the liver,and improves the abnormal lipid metabolism in the liver caused by OVX.
