摘要:目的:观察贞芪扶正颗粒联合奥沙利铂对H_(22)移植瘤的抑制效果并探讨肿瘤相关髓样分化因子(Myeloid differentiation factor 88,MyD88)、核因子-κB(Nuclear Factor-κB p65,NF-κB p65)、肿瘤坏死因子(Tumor necrosis factor-α,TNF-α)的调节机制。方法:72只KM小鼠随机分为空白对照组、模型对照组、奥沙利铂10 mg/kg组、贞芪扶正颗粒0.026、0.013、0.007 g/kg+奥沙利铂10 mg/kg组,建立H_(22)荷瘤小鼠模型,经药物干预10 d后,剥取瘤体,进行瘤体测量、称重和抑瘤率分析;HE染色法观察瘤体组织形态学变化;Elisa法测定血清中TNF-α蛋白含量;WB和qPCR法分别检测瘤体中MyD88、NF-κB p65蛋白和mRNA水平。结果:与奥沙利铂10 mg/kg组相比,奥沙利铂10 mg/kg+贞芪扶正颗粒0.026、0.013、0.007 g/kg组瘤体体积和体质量均明显减小(P<0.05或P<0.01),抑瘤率上升;HE染色观察发现各治疗组坏死肿瘤细胞增多、细胞核碎裂并消失,其中0.026 g/kg组可见纤维组织增生;血清中TNF-α水平显著增高(P<0.01),瘤体组织中MyD88、NF-κB p65蛋白和mRNA水平显著下调(P<0.05),且呈现贞芪扶正颗粒浓度剂量依赖性关系。结论:贞芪扶正颗粒具有促进奥沙利铂抑制H_(22)移植瘤增长的作用,其分子机制可能与调节MyD88、NF-κB p65和TNF-α水平有关。
Objective:To observe the inhibitory effect of Zhenqifuzheng granule combined with Oxaliplatin on H_(22)-bearing mice and explore the regulatory mechanism of MyD88,NF-κB p65 and TNF-α.Methods:72 KM mice were randomly divided into:the cotrol group,the model group,10 mg/kg Oxaliplatin group,Zhenqifuzheng granule combined with 10 mg/kg Oxaliplatin groups(0.026,0.013,0.007 g/kg).H_(22)-bearing mice model was established,10 days after treatment,the tumors were dissected,measured and weighed to calculated the inhibitory rate.The histological changes of the tumor were observed by HE.TNF-αcontent in serum was measured by ELISA.The levels of MyD88,NF-κB p65 protein and mRNA in the tumor tissues were detected by WB and qPCR.Results:Compared with 10 mg/kg Oxaliplatin group,the volumes and weights of tumors were decreased in all doses of Zhenqifuzheng granule combined with oxaliplatin groups(0.026,0.013,0.007 g/kg)(P<0.05 or P<0.01)),while the inhibitory rate was increased.HE results showed that necrotic tumor cells were increased,the nucleus disintegrated and disappeared in all doses of combination groups,the fibrous tissue hyperplasia was observed in 0.026 g/kg combination group.The serum level of TNF-αwas significantly increased(P<0.01),the contents of MyD88,NF-κB p65 protein and mRNA in tumor tissues were significantly decreased(P<0.05),in concentration-dose-dependent manner.Conclusions:Zhenqifuzheng granule can promote Oxaliplatin to inhibit the growth of H_(22)cancer cell,its molecular mechanism may be related to regulating the levels.of MyD88,NF-κB p65 and TNF-α.
