摘要:2013年在甘肃省陇西县发现一种严重为害药用作物玉竹(Polygonatum odoratum)叶部的病害,症状类似于其他作物的褐斑病,但病原不明。采集发病叶片,分离培养病原菌并镜检观察其形态,进行ITS序列扩增及分析,按照科赫氏法则测定其致病性。经过分离培养和纯化后,获得菌株Ps01、Ps02、Ps03,通过回接试验确定Ps02为致病菌;结合形态学特征和ITS序列分析,将陇西的玉竹褐斑病病原鉴定为交链格孢菌(Alternaria alternata)。
In 2013, a serious disease was found on the leaves of the medical plant Polygonatum odoratum in Longxi of Gansu Province, with symptoms similar to brown spot on other crops, but the pathogen was unknown. To determine the pathogen, we isolated and cultured the pathogens from infected leaves and examined their morphology by microscopic observation, then the ITS sequences were amplified and analyzed, and the pathogenicity was determined according to Koch′ s postulates.Strains Ps01, Ps02, and Ps03 were obtained after isolation, culture, and purification of the pathogens, and Ps02 was determined to be the pathogen of the disease by re-inoculation test. Based on morphological characteristics and ITS sequence analysis, the pathogen of the brown spot of Polygonatum odoratum in Longxi was identified as Alternaria alternata.
