摘要:目的 探究炙红芪对脾气虚模型大鼠肠道菌群组成及短链脂肪酸代谢的影响。方法 用疲劳、限制饮食、泻下三因素复合造模方法进行脾气虚模型大鼠复制,并随机分为模型组、实验组,每组15只;另取15只正常SD大鼠作为空白组。实验组灌胃给予12.6 g·kg^(-1)炙红芪水提液,空白组和模型组均灌胃给予等剂量的蒸馏水。用16SrRNA检测大鼠肠道菌群组成;用GC-MS法检测结肠内容物短链脂肪酸含量,并进行差异菌属筛选及相关性分析。结果 空白组、模型组和实验组的Ruminococcaceae_UCG-005相对丰度分别为36.32±9.31×10^(-3)、3.58±1.37×10^(-3)和32.77±6.67×10^(-3),g_Bifidobacterium相对丰度分别为4.60±5.46×10^(-3)、91.78±4.30×10^(-2)和12.07±9.83×10^(-3),g_Faecalibaculum相对丰度分别为0.78±0.30×10^(-3)、43.80±5.47×10^(-3)和16.94±1.88×10^(-3),总短链脂肪酸含量分别为(21.98±1.85)、(10.48±3.17)和(17.26±3.10) mg·g^(-1)。模型组的上述指标与实验组和空白组比较,差异均有统计学意义(P<0.05,P<0.01)。差异菌属与短链脂肪酸有一定的相关性。结论 炙红芪可能通过调节肠道菌群,改善短链脂肪酸代谢起到治疗脾气虚大鼠作用。
Objective To explore the effect of Hedysari Radix Preparata cum Melle on intestinal flora composition and short-chain fatty acid metabolism of spleen-qi deficiency model rats.Methods The spleen-qi deficiency model rats were constructed by using the compound modeling method including fatigue,diet restriction and diarrhea,and they were randomly divided into model and experimental group with 15 cases per group;another 15 normal SD rats were taken as blank group.Experimental group was given 12.6 g·kg^(-1)Hedysari Radix water by gavage.Blank and model groups were given the same dose of distilled water by gavage.16SrRNA and GC-MS methods were employed to detect the composition of intestinal microflora and the contents of short-chain fatty acids in colon contents,respectively.Also,the differential bacteria were screened and correlation analysis was performed.Results The relative abundances of Ruminococcaceae_UCG-005 in the blank,model and experimental groups were 36.32±9.31×10^(-3),3.58±1.37×10^(-3) and 32.77±6.67×10^(-3);the relative abundances of g_Bifidobacterium were 4.60±5.46×10^(-3),91.78±4.30×10^(-2) and 12.07±9.83×10^(-3);the relative abundances of g_Faecalibaculum were 0.78±0.30×10^(-3),43.80±5.47×10^(-3) and 16.94±1.88×10^(-3);total short-chain faty acid contents were(21.98±1.85),(10.48±3.17)and(17.26±3.10)mg:g=1.The differences of above indexes were statistically significant between the model and the blank and experimental groups(P<0.05,P<0.01).The results showed that there was a certain correlation between differential bacteria and short-chain fatty acids.Conclusion The Hedysari Radix Preparata cum Melle may play an important role in treating spleen-qi deficiency rats by regulating intestinal flora and improving short-chain fatty acids metabolism.
