摘要:目的:研究IκB激酶/核因子κB/环氧酶2(IKKα/NF-κB/COX-2)信号通路分子在溃疡性结肠炎(UC)大鼠结肠中的表达及乌梅丸的干预作用,探讨乌梅丸治疗UC的作用机制并验证网络药理学结果。方法:采用2,4,6-三硝基苯磺酸(TNBS)/乙醇灌肠复制UC大鼠模型,在评价疾病活动指数(DAI)、病理组织学评分等基础上,采用实时荧光定量聚合酶链反应技术(RT-PCR)检测结肠组织Ikkα mRNA、Nf-κb mRNA、Cox-2 mRNA表达,蛋白质免疫印迹技术(WB)测定结肠组织IKKα、NF-κB、COX-2蛋白表达。结果:TNBS/乙醇诱导的UC模型对照组大鼠DAI、结肠组织病理学评分增高,大鼠结肠组织IKKα、NF-κB、COX-2蛋白及mRNA表达显著上调(P<0.01)。乌梅丸28.8、14.4 g/kg可显著降低UC模型对照组大鼠DAI和结肠组织病理学评分(P<0.01),下调结肠组织IKKα、NF-κB、COX-2蛋白及mRNA的表达(P<0.05或P<0.01)。结论:下调UC模型对照组大鼠结肠组织IKKα/NF-κB/COX-2信号传导通路,抑制炎症反应可能是乌梅丸治疗UC的重要机制之一。
Objective: To investigate the expression of IKKα/NF-κB/COX-2 signaling pathway in UC rats and the effects of Wumeiwan, the research on the potential mechanism of Wumeiwan treating ulcerative colitis(UC) and to verify its network pharmacological result.Methods: UC rats model was induced by enema with 2,4,6-trinitrobenzene sulfonic acid(TNBS)/ethanol. The disease activity index(DAI) and histopathological score were evaluated. The levels of IKKα mRNA, NF-κB mRNA and COX-2 mRNA were measured by real-time fluorescence quantification polymerase chain reaction( RT-PCR). The levels of IKKα, NF-κB and COX-2 were measured by western blotting(WB).Results: Compared with the control group, the levels of DAI and histopathological score were improved in UC model group, the levels and protein expressions of IKKα mRNA, NF-κB mRNA and COX-2 mRNA were significantly up regulated(P<0.01). Compared with the model group, the levels of DAI and histopathological score were reduced in 14.4 g/kg and 28.8 g/kg Wumeiwan decoction groups, the levels and protein expressions of IKKα mRNA, NF-κB mRNA and COX-2 mRNA were significantly down regulated(P<0.05 orP<0.01). Conclusion: Wumeiwan has certain therapeutic effect on UC in rats, the mechanism may be related to down regulating the IKKα/NF-κB/COX-2 signaling pathways in colon and inhibiting the inflammatory reaction.
