摘要:目的:探讨解毒化浊方对胰岛素抵抗大鼠葡萄糖转运4(glucose transporter 4,GLUT4)表达的影响及其治疗胰岛素抵抗的作用机理。方法:将雄性wistar大鼠48只随机选出12只设为空白组,采用生理盐水灌胃,其余大鼠采用高脂饲料喂养方法造胰岛素抵抗大鼠模型后,随机分为模型组、解毒化浊治疗组及罗格列酮治疗组,每组12只,解毒化浊组以解毒化浊方混悬液2.025g/(kg·d)灌胃,罗格列酮组以罗格列酮混悬液0.36 mg/(kg·d)灌胃,模型组与空白对照组均予等体积生理盐水灌胃。治疗10周后检测大鼠空腹血糖(FBG)、糖耐量试验(2 h血糖)[OGTT(2h PG)]、空腹胰岛素(FINS)水平,并应用反转录聚合酶链式反应(RT-PCR)方法检测各组大鼠骨骼肌组织GLUT4 mRNA的水平。结果:解毒化浊方与罗格列酮均能降低大鼠FBG、2h PG及FINS水平,差异有统计学意义(P<0.05),在降低体质量方面与空白组比较解毒化浊组及罗格列酮组差异有统计学意义(P<0.05);解毒化浊组与罗格列酮组大鼠血清FBG、2h PG、FINS比较差异无统计学意义(P>0.05);与空白组比较,模型组大鼠GLUT4表达水平呈低表达(P<0.05);与模型组比较,解毒化浊组与罗格列酮组大鼠GLUT4表达水平显著增加(P<0.05)。结论:解毒化浊方可降低胰岛素抵抗大鼠血清FBG、OGTT(2h PG)及FINS水平,增加骨骼肌组织中GLUT4的表达,提高胰岛素敏感性,有改善胰岛素抵抗的作用。
Objective: To explore the impact ofJieDu HuaZhuo prescription on the expressions of glucose transporter 4 (GLUT4) mRNA of the rats with insulin resistance(IR). Methods: Twelve rats were chosen from 48 male Wistar ones, established as the blank group and accepted intragastric administration of physiological saline, the rest rats were fed with high lipid fodder to prepare IR rat model and randomly divided into the model group, the treatment group ofJieDu HuaZhuo prescription and the treatment group of rosiglitazone, 12 rats each group, the treatment group ofJieDu HuaZhuo prescription received the lavage ofJieDu HuaZhuo suspension in the concentration of2.025g/(kg. d) and the treatment group of rosiglitazone rosiglitazone suspension in the concentration of 0.36mg/(kg.d), the model group and blank control group equivalent volumes of physiological saline lavage. To detect the levels of FBG, OGTT (2hPG) and FINS of the rats after treating for ten weeks, and measure the levels of GLUT4mRNA in skeletal muscles of the rats by RT-PCR. Results: JieDu HuaZhuo prescription and rosiglitazone could decrease the levels of FBG, 2hPG and FINS of the rats, the difference showed statistical meaning(P〈0.05), the difference had statistical meaning in reducing body weight within the treatment group ofJieDu HuaZhuo prescription(P〈0.05); the difference presented no statistical meaning in the levels of FBG, 2hPG and FINS between the treatment groups (P〉0.05); compared with the blank group, the levels of GLUT4 expressions in the model group were low (P〈0.05); compared with the model group, the expressions of GLUT4 in the treatment groups increased significantly (P〈0.05). Conclusion: JieDu HuaZhuo prescription could decrease the levels ofFBG, 2hPG and FINS of IR rats, enhance GLUT4 expressions in the skeletal muscle and improve insulin sensitivity, therefore, to improve IR.
