摘要:目的:考察油橄榄叶提取物对3种模型动物的降血糖、降血脂作用并探讨相关机制。方法:采用db/db自发性糖尿病小鼠作为主要研究对象,通过监测给药后空腹血糖(FBG)、葡萄糖耐量(OGTT)、胰岛素耐量(ITT)、胰岛素水平(INS)、胰岛素敏感指数(ISI)、糖化血清蛋白(GSP)及游离脂肪酸(NEFA),考察油橄榄叶提取物250、500、1000 mg/kg对2型糖尿病db/db模型小鼠的降血糖作用;同时还考察油橄榄叶提取物125、250及500 mg/kg对肾上腺素诱导SD大鼠的降血糖作用以及油橄榄叶提取物100 mg/kg、300 mg/kg对ApoE基因敲除小鼠脂质代谢的作用。结果:与正常对照组比较,模型对照组血糖、糖耐量、胰岛素耐量显著升高(P<0.01),糖化血清蛋白含量、血清胰岛素水平、游离脂肪酸含量显著升高,胰岛素敏感指数显著降低。油橄榄叶提取物500 mg/kg、1000 mg/kg可显著降低db/db小鼠的空腹血糖(P<0.05或P<0.01);250、500、1000 mg/kg剂量组可显著降低GSP、NEFA水平(P<0.01),显著升高ISI(P<0.05或P<0.01);油橄榄叶提取物125、250、500 mg/kg可显著降低肾上腺素SD大鼠的血糖水平(P<0.05或P<0.01);油橄榄叶提取物100、300 mg/kg还可显著降低ApoE基因敲除小鼠的三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)及超低密度脂蛋白胆固醇(VLDL-C)水平(P<0.05或P<0.01)。结论:油橄榄叶提取物对db/db自发性糖尿病模型小鼠、肾上腺素SD大鼠具有降血糖作用,对ApoE基因敲除模型小鼠具有降脂作用,其作用机制可能与调节脂质代谢、改善胰岛素抵抗有关,也可能与其能够抑制糖原动员、糖异生有关,其作用机制有待进一步深入研究。
Objective:To investigate the hypoglycemic and hypolipidemic effects of Olea europaea L.leaves extract(OLE)on 3 animal models and its possible mechanism.Methods:The db/db spontaneous diabetes mice were taken as experimental subject,the levels of fast blood glucose(FBG),oral glucose tolerance test(OGTT),insulin tolerance test(ITT),insulin(INS),insulin sensitivity index(ISI),glycated serum protein(GSP)and non-esterified fatty acid(NEFA)were observed after administration to explore the hypoglycemic effect of different doses of OLE(250,500,1000 mg/kg).Meanwhile,the hypoglycemic effect of different doses of OLE(125,250 and 500 mg/kg)on hyperglycemia SD rat induced by epinephrine was investigated.Moreover,the antihyperlipidemic effect of OLE(100 mg/kg、300 mg/kg)in ApoE gene knockout mice were observed.Results:Compared with the control group,the levels of FBG,OGTT and ITT were significantly increased(P<0.01),the levels of GSP,INS and NEFA were increased,the ISI was decreased.Compared with the model group,500 and 1000 mg/kg OLE significantly decreased the FBG(P<0.05,P<0.01),250,500 and 1000 mg/kg OLE significantly decreased the levels of GSP and NEFA(P<0.01)and increased the ISI level(P<0.05,P<0.01)of db/db mice.In the hyperglycemia SD rats induced by epinephrine,the blood glucose levels in OLE groups(125,250 and 500 mg/kg)were significantly decreased(P<0.05,P<0.01).The serum levels of TG,TC,LDL-C and VLDL-C of ApoE mice were significantly decreased in OLE groups(100,300 mg/kg)(P<0.05,P<0.01).Conclusion:OLE shows a hypoglycemic effect on spontaneous type 2 diabetes db/db mice and hyperglycemia SD rats induced by epinephrine,it also shows a hypolipidemic effect on ApoE gene knockout mice.The mechanism may be related to regulation of lipid metabolism and insulin resistant,or the inhibition of glycogen mobilization and gluconeogenesis,which needs to be further studied.
