摘要:目的制备湿生扁蕾乙酸乙酯提取物并探讨其对RAW246.7细胞免疫调节的影响。方法采用正交实验设计优化提取工艺,采用乙酸乙酯萃取分离得提取物,采用HPLC法检测提取物中有效成分当药黄素、木犀草素、当药宁的含量。以RAW246.7细胞为研究对象,分别设置空白组、模型组、乙酸乙酯提取物高、中、低剂量组,检测细胞活率、细胞吞噬率、M1型极化细胞比率、白介素-6(IL-6)和IL-1β的含量及肿瘤坏死因子α(TNF-α)和IL-10表达。结果湿生扁蕾乙酸乙酯提取物的最佳制备工艺为:95%乙醇提取3次,料液比1:12,提取时间1.5 h,乙酸乙酯萃取4次。提取物可显著抑制RAW246.7细胞M1型极化,并降低IL-6和IL-1β的含量,抑制TNF-α蛋白表达,上调IL-10蛋白表达,提高细胞吞噬能力。结论湿生扁蕾乙酸乙酯提取物可抑制RAW246.7巨噬细胞M1型极化,改善细胞免疫调节活性。
OBJECTIVE To prepare ethyl acetate extract of Gentianopsis paludosa and explore its effect on the immune regulation of RAW246.7 cells.METHODS Orthogonal design was used to optimize thel extraction process for G.paludosa.The extract of G.paludosa was separated and purified using the ethyl acetate extraction method.HPLC was used to detect the effective components,including swertisin,luteolin,and swertianine.Using RAW246.7 cells as the research object,cells were divided into control group,model group,and high,medium,and low dose groups of ethyl acetate extract from G.paludosa.Cell viability,phagocytosis rate,M1 polarization cell ratio,cytokines IL-6 and IL-1βcontents as well as TNF-αand IL-10 protein expression were determined.RESULTS The optimal preparation process for G.paludosa was to extract three times with 95%ethanol,a solid-liquid ratio of 1∶12,an extraction time of 1.5 h,and to extract 4 times with ethyl acetate.The extract of ethyl acetate of G.paludosa significantly inhibited M1 polarization in RAW246.7 cells,reduced IL-6 and IL-1βlevels,downregulated TNF-αprotein expression and upregulated IL-10 protein expression.CONCLUSION The extraction of ethyl acetate from G.paludosa inhibited M1-type polarization of RAW246.7 macrophages and improved the immunomodulatory activity.
