摘要:目的 :探讨红芪多糖 (HPS)对糖尿病 (DM)大鼠肾脏保护作用的机制。方法 :实验采用随机方法。左下腹腔注射链脲菌素 (STZ) 6 5 mg/ kg制备 DM大鼠模型 ,以 72 h后空腹血糖 >16 .7mm ol/ L 为模型成功标志。将 DM大鼠随机分为模型组、依那普利组及 HPS大、小剂量组。依那普利灌胃剂量为 7mg/ kg,HPS灌胃剂量为 15 0和 5 0 mg/ kg,余灌胃等量生理盐水 ,均每日 1次 ,连续 8周。灌胃 2周和 8周末次给药后禁食 12 h(不禁水 ) ,测血糖 ;分两批处死动物 ,取肾进行组织匀浆 ,测定一氧化氮 (NO)、一氧化氮合酶 (NOS)及超氧化物歧化酶 (SOD)、丙二醛 (MDA)等指标。结果 :HPS治疗后 DM大鼠血糖显著降低 ,且随治疗时间的延长 ,降糖作用逐渐明显。DM大鼠肾组织 NO和 NOS活性呈明显的早期升高、晚期下降趋势 ;使用 HPS2周时显著抑制了NO和 NOS活性的过分升高 ,8周时显著抑制了 NO和 NOS活性的持续降低。随病程延长 DM大鼠肾组织SOD活性呈逐渐下降、MDA呈逐渐升高的趋势 ;HPS治疗后肾组织 SOD活性明显增强 ,8周时有显著下降 ;MDA则呈下降趋势。以上作用大剂量 HPS组均优于小剂量组 ,呈明显的量效关系 ;依那普利组作用不明显。结论 :HPS能有效降低 DM大鼠血糖 ,改善和调节肾组织过氧化反应指标 ,可能对 DM肾脏损害有保护?
Objective: To investigate the protective effect of hedysari polybotrys polysaccharide(HPS,红芪多糖) on kidney in diabetic rats. Methods: Sixteen rats were selected randomly for normal control rats and injected equal volume of cusion liquid into leftlower part of abdominal cavity. Others were injected streptozotocin(STZ) 65 mg/kg into leftlower abdominal cavity and measured the blood fasting sugar after 72 hours. The rats, whose consistency of blood fasting sugar were onto 16 7 mmol/L, were randomly divided into following groups: model group, STZ diabetic rats treated with enalapril, diabetic rats treated with large dosage of HPS, diabetic rats treated with low dosage of HPS. All groups were killed while they were on diet after 12 hours at 2 th and 8 th weekend. Then the relevant indexes in tissue fluid of kidney were measured. Results: It presented that the activity of nitric oxide(NO) and nitric oxide synthase(NOS) rised at early stage and descented at later period obviously in the STZ diabetic rats. The activity of NO and NOS in diabetic rats treated with large dosage of HPS were significantly lower than diabetic rats untreated group of early stage(P<0 01)and higher of later period(P>0 01). The activity of superoxide dismutase(SOD) in tissue fluid of kidney was improved, and the content of malondialdehyde(MDA) in the tissue fluid of kidney was kept decreasing in diabetic rats treated with HPS. Conclusion: HPS can cut down the blood sugar of diabetic rats and perfect the relevant indexes in tissue fluid of kidney. HPS has some protective effect on kidney of diabetic rat.
