摘要:目的研究肺癌细胞A549条件培养液诱导间充质干细胞(MSC)在增殖、迁移、成瘤及侵袭方面的改变。方法设MSC为对照组,A549细胞条件培养液培养的MSC为模型组(T-MSC)。通过检测细胞形态、细胞增殖及迁移能力评估各组MSC发生恶性转化的程度;经Westernblot检测各组MSC恶性分化相关NF-κB及STAT3的活性;通过体内实验研究各组MSC的成瘤能力。结果T-MSC组在形态上出现改变,从纤维样漩涡状改变为椭圆形纺锤状;T-MSC组增殖能力和体外迁移能力高于对照组MSC(P<0.05);T-MSC组p65及STAT3蛋白磷酸化水平高于对照组MSC(P<0.05);T-MSC组细胞可在NOD-SCID小鼠皮下出现成瘤样表型且实体瘤表现侵袭浸润样表型。结论A549细胞条件培养液可诱导MSC发生一定程度的恶性分化,表现出细胞形态学改变、增殖及迁移能力提高、获得成瘤样表型,这可能是通过调控NF-κB及STAT3活性来实现的。
Objective Study the effects of conditional-medium from lung cancer cells A549 on proliferation,migration,tumorigenicity and invation of mesenchymal stem cells(MSC).Methods Grouping MSC as control group and A549 cells-conditioned medium-cultured MSC as model group(T-MSC).Evaluating the degree of malignant transformation in each group by detecting cell morphology,proliferation and migration.The malignant differentiation-associated NF-κB and STAT3 activities were detected by Western blot analysis in each group.The tumorigenicity of each group was detected in vivo.Results Cells in T-MSC group exert morphological changes fromfibrously swirls shape to elliptically spindle-like shape.Cells in T-MSC group exert higher proliferation and migration compared with cells in control group(P<0.05).Phosphorylation level of p65 and STAT3 in T-MSC group are higher than those in control group(P<0.05).Cells in T-MSC group exert tumorigenic phenotype and invasive and infiltration-like phenotype in NOD-SCID mice.Conclusions MSC cultured in A549 cells-conditioned medium exert malignant transformation in some degree,displaying morphological change,elevated proliferation and migration,tumor-like phenotype,which may be fulfilled though activating NF-κB and STAT3 in T-MSC.
