摘要:目的:探讨敦煌医方小补肾汤对镉染毒大鼠骨和肾功能的影响。方法:将SPF级Wistar大鼠60只按体质量随机分为6组:空白组、乙酰半胱氨酸组、模型组及小补肾汤高、中、低剂量组。空白组腹腔注射0.9%Na Cl,其余5组均腹腔注射0.1%氯化镉,1.5 mg/kg注射,相当于1.5 m L/kg,每周5次,共5周,进行染毒。染毒的同时,分别给予相应干预,乙酰半胱氨酸组组予乙酰半胱氨酸泡腾片溶液(5.4 mg/m L)1 m L/100 g,小补肾汤各剂量组予相应剂量(高:2.7 g/m L,中:1.35 g/m L,低:0.675 g/m L)小补肾汤1 m L/100 g灌胃治疗,每天1次,连续5周。最后1次注射后,取血检测。结果:与空白对照组比较,模型组大鼠血清钙、磷、骨钙素(OC)显著降低,2组比较差异有统计学意义(P<0.05);与模型组比较,乙酰半胱氨酸组、小补肾汤高中低剂量组大鼠血清钙、磷、OC显著增加,差异有统计学意义(P<0.05)。与空白对照组比较,模型组大鼠血清碱性磷酸酶(ALP)、抗酒石酸酸性磷酸酶5b(TRACP-5b)、尿素氮(BUN)显著升高,2组比较差异有统计学意义(P<0.05);与模型组比较,乙酰半胱氨酸组、小补肾汤高中低剂量组大鼠血清ALP、TRACP-5b、BUN显著降低,差异有统计学意义(P<0.05)。结论:小补肾汤对于镉染毒大鼠骨和肾功能具有一定的修复作用。
Objective: To discuss the effects of Dunhuang prescription Xiao BuShen Tang on bone and renal functions of the rats exposed to cadmium. Methods: Sixty rats were randomly divided into six groups: the blank group, acetylcysteine group, the model group, the groups of high, moderate and low dosages of Xiao BuShen Tang. The blank group accepted peritoneal injection of 0.9% NaCl, other groups were given intraperitoneal injection of 0.1% cadmium chloride, 1.5 mg/kg injection, equivalent to 1.5 mL/kg, five times each week, a total of five weeks. At the same time, they accepted corresponding intervention measures respectively, acetylcysteine group received acetylcysteine effervescent tablets solution (5.4 mg/mL) 1 mL / 100 g, different dosages groups ofXiao BuShen Tang accepted intragastric administration of different dosages (high: 2.7g/mL, moderate: 1.35g/mL and low: 0.675g/mL) ofXiao BuShen Tang in the density of 1mL/100g, once per day, for five weeks consecutively. The blood was tested after the last injection. Results: Compared with blank control group, the levels of serum calcium, phosphorus and osteocalcin (OC) of the model group decreased notably, the difference showed statistical meaning between both groups (P〈0.05); compared with the model group, the levels of serum calcium, phosphorus and OC increased in acetylcysteine group, high, moderate and low dosages groups ofXiao BuShen Tang significantly, and the difference showed statistical meaning (P〈0.05). Compared with blank control group, the levels of ALP, TRACP-5b and BUN elevated remarkably in the model group, and the difference had statistical meaning (P〈0.05); compared with the model group, the levels of ALP, TRACP-5b and BUN decreased remarkably in acetylcysteine group, high, moderate and low dosages groups ofXiao BuShen Tang, and the difference showed statistical meaning (P〈0.05). Conclusion: Xiao BuShen Tang could recover bone and renal functions of the rats suffering cadmium exposure to a certain degree.
